Chemoprevention of benzene-induced bone marrow and pulmonary genotoxicity

Abstract: Our earlier studies documented the ability of 7,8-diacetoxy-4-methylcoumarin (DAMC) to cause irreversible inhibition of cytochrome P-450 linked mixed function oxidases (MFO) mediated by membrane bound DAMC: protein transacetylase. Since P-450 catalyzed oxidation of benzene is crucial to its toxic effects, the action of DAMC and related analogues were considered promising in preventing the genotoxicity due to benzene. For this purpose rats were pretreated with various acetoxy-4-methylcoumarins (test compounds),… Show more

“…By the method of flow cytometry or by histochemical staining, induction of apoptosis by aflatoxin B was reported in rat hepatocytes (Meki et al, 2001), lung cells and bone marrow cells (Raj et al, 2001), or human bronchial epithelial cells (X-J Yang et al, 2012). In the present study, the percentages of apoptotic cells in the three immune organs were all higher than those of control group by flow cytometry, and apoptotic cells and typical apoptotic bodies could be observed in the thymus, BF and spleen by electron microscopy.…”

“…Our perception of acetyl CoA‐independent acetylation of proteins had its origin in our investigations on the mechanism of biochemical action of DAMC. We had earlier demonstrated the remarkable inhibitory action of DAMC on several microsomal cytochrome P ‐450‐linked mixed function oxidases (MFO) catalyzing aflatoxin B 1 ‐epoxidation, dealkylation of alkylated resorufins [6,10] and toxicokinetics of benzene [11]. The irreversible inhibition of MFO mediated by DAMC was in contrast to the action of classical inhibitors of cytochrome P ‐450 [12–15] in that DAMC needed no prior oxidative metabolism.…”
