Chemokines Regulate IL-6 and IL-8 Production by Fibroblast-Like Synoviocytes from Patients with Rheumatoid Arthritis

Abstract: Rheumatoid arthritis (RA) is characterized by proliferation of synoviocytes that produce inflammatory cytokines and chemokines. The expressed chemokines are thought to be involved in the migration of inflammatory cells into the synovium. In this study we show that CCL2/monocyte chemotactic protein-1, CCL5/RANTES, and CXCL12/stromal cell-derived factor-1 enhanced IL-6 and IL-8 production by fibroblast-like synoviocytes (FLS) from patients with RA, and their corresponding receptors, CCR2, CCR5, and CXCR4, respec… Show more

“…Conversely, chemokines may also stimulate the release of proinflammatory cytokines. For instance, CCL3 can stimulate the synthesis of IL-1, IL-6, and TNF␣ (26), and CCL5 can induce the production of IL-6 and IL-8 (27).…”

“…There are several other reports that suggest the relevance of MAPK activation to the immunopathogenesis of RA (48)(49)(50)(51)(52). However, no report has described the effects of MAPK pathway inhibitors on the ex vivo cytokine production by freshly isolated synovial cells from RA patients.…”

“…For example, skin, bone marrow, and RA synovial fluid fibroblasts all spontaneously make IL-6 [110][111][112]. Increased levels of TNF-α, IL-6, IFN-γ [113] and IL-17A [114] were found when synovial fibroblasts were cultivated with T cells.…”

“…For instance, CCL2 (monocyte chemotactic protein-1 or MCP-1), CCL5 (RANTES), and CXCL12 (SDF-1) have all been shown to enhance IL-6 and IL-8 production by RA synovial fibroblasts [111]. IL-8 is a chemoattractant for neutrophils and thus a regulator of acute, innate inflammation, but also a promoter of angiogenesis.…”
