Chemokines and Growth Factors Produced by Lymphocytes in the Incompetent Great Saphenous Vein

Abstract: The role of cytokines in the pathogenesis of chronic venous disease (CVD) remains obscure. It has been postulated that oscillatory flow present in incompetent veins causes proinflammatory changes. Our earlier study confirmed this hypothesis. This study is aimed at assessing chemokines and growth factors (GFs) released by lymphocytes in patients with great saphenous vein (GSV) incompetence. In 34 patients exhibiting reflux in GSV, blood was derived from the cubital vein and from the incompetent saphenofemoral j… Show more

“…Tissue hypoxia leads to apoptosis and extracellular changes [ 129 ]. Aging-induced inflammation in veins is associated with elevated cytokine production [ 130 ] and increased MMP activation, which likely play a critical role in the pathogenesis of chronic venous disease [ 131 , 132 ].…”

“…Then, after the last washing, the beads are suspended in a buffer for further analysis. Washing was performed using an ELx 50 magnetic washer (BioTek, Shoreline, WA, USA) and quantitative measurements of cytokines were analyzed using the Bio-Plex 200 System controlled by Bio-Plex Manager software (Standard Edition, Bio-Rad Laboratories Inc., Hercules, CA, USA) [ 17 , 18 , 19 , 20 , 21 ].…”

“…The intra-assay %CV varied up to 15%, and the inter-assay %CV varied up to 25%, depending on the analyzed cytokine. The method has been previously used in measuring cytokines in serum and supernatants [16,17].…”
