Chemokine and cytokine expression in murine intestinal epithelium following Nippostrongylus brasiliensis infection

Abstract: Infection of mice with the nematode parasite Nippostrongylus brasiliensis results in a well characterized intestinal mastocytosis with intraepithelial migration of mucosal mast cells (MMC). The molecules mediating this response are unknown. We examined expression of several putative mast cell chemoattractants in intestinal epithelium following N. brasiliensis infection. Expression of the chemokines monocyte chemoattractant protein-1 (MCP-1), macrophage inflammatory protein-1 alpha (MIP-1alpha), RANTES (regulat… Show more

“…Subsequent purification and removal of contaminating DNA using DNA-free DNase (Ambion, Austin, TX) has been previously described (17). To check the purity of stripped epithelium preparations, samples were simultaneously fixed in 4% parafomaldehyde for embedding, sectioning, and H&E staining as previously described (19). Tissue samples for total RNA extraction were also collected from nonperfused uninfected and infected female BALB/c mice day 14 following T. spiralis infection.…”

“…To check infections were successful, adult worms were isolated from groups of four to five of the mice at 6 -7 days after infection as previously described (17). For RT-PCR analysis, mice were killed on day 14 following infection (worm expulsion phase) and epithelium exfoliated by EDTA perfusion as previously described (18,19). Samples were also prepared from age-matched uninfected controls.…”

“…Each PCR was conducted in a total volume of 25 l containing 12.5 l of master mix (Promega) and 2.5 l of primer mix (10 M). GAPDH (35), IFN-␥, and IL-4 (36) primers were obtained from ThermoHybaid (Ulm, Germany). Primers for IFN-␥ and IL-4 were intron-spanning to allow discrimination of any genomic DNA.…”

“…Adhesion of CMC grown in a combination of IL-3, SCF, and IL-9, all of which are expressed in the gut (29,30) and are known mast cell growth factors (31-33), has not previously been studied. As shown above, CMC grown in the absence of TGF-␤1 did not adhere to laminin-1.…”
