Chemogenetic manipulation of astrocytic signaling in the basolateral amygdala reduces binge‐like alcohol consumption in male mice

Abstract: Binge drinking is a common occurrence in the United States, but a high concentration of alcohol in the blood has been shown to have reinforcing and reciprocal effects on the neuroimmune system in both dependent and non-dependent scenarios. The first part of this study examined alcohol's effects on the astrocytic response in the central amygdala and basolateral amygdala (BLA) in a non-dependent model. C57BL/6J mice

“…Research showed that in vivo activation of Gq‐DREADDs, expressed in astrocytes of the nucleus accumbens core, modulated activity of the surrounding neuropil via release of glutamate (Scofield et al, 2015). Furthermore, glutamate concentrations were elevated in the amygdala after activation of Gq‐DREADDs expressed on astrocytes, which in turn reversed the reduced glutamate levels after ethanol consumption in mice (Nwachukwu et al, 2021). Likewise, activation of Gq‐DREADDs expressed in astrocytes of hippocampal and striatal brain slices was proposed to elicit glutamate release (Chai et al, 2017; Durkee et al, 2019).…”

“…Esta técnica se utiliza para modificar la actividad astrocitaria que en general resulta en cambios en la señalización del Ca 2+ y diversos efectos en otras células cerebrales (45,46). La principal ventaja de esta técnica es la activación o inhibición específica de los astrocitos frente al enfoque farmacológico clásico que afecta de manera menos específica pudiendo alterar la función neuronal y/o producir diferentes efectos secundarios (47)(48)(49)(50).…”

“…Activation of PFC astrocytes increased ethanol consumption in ethanol-naïve male mice whereas reduction of astrocyte calcium via CalEx expression reduced ethanol drinking indicating a bidirectional regulation of ethanol-consumption by PFC astrocytes ( Erickson et al, 2021 ). In the basolateral amygdala, activation of astrocytes reduced ethanol consumption in male mice ( Nwachukwu et al, 2021 ). Likewise, in the nucleus accumbens (NAc), a key reward center, activation of astrocytes in male rats decreased ethanol-seeking behavior after abstinence ( Bull et al, 2014 ).…”
