Chemistry of the Strong Electrophilic Metal Fragment [99Tc(N)(PXP)]2+ (PXP = Diphosphine Ligand). A Novel Tool for the Selective Labeling of Small Molecules

Abstract: Monosubstituted [M(N)Cl(2)(POP)] [M = Tc, 1; Re, 2] and [M(N)Cl(2)(PNP)] [M = Tc, 3; Re, 4] complexes were prepared by reaction of the precursors [M(N)Cl(4)](-) and [M(N)Cl(2)(PPh(3))(2)] (M = Tc, Re) with the diphosphine ligands bis(2-diphenylphosphinoethyl)ether (POP) and bis(2-diphenylphosphinoethyl)methoxyethylamine (PNP) in refluxing dichloromethane/methanol solutions. In these compounds, the diphosphine acted as a chelating ligand bound to the metal center through the two phosphorus atoms. Considering al… Show more

The methylene carbons of the diphosphane chain, C(1), C(4), C(2), and C(3), are located, respectively, at 1.69, 1.81, 2.64 and 2.64 Å below the mean S 2 P 2 basal plane, whereas N(2) faces this plane at the intermediate distance of 2.29 Å .…”

“…Bolzati et al investigated a novel series of cationic 99m Tc-nitrido complexes containing a [ 99m Tc ≡ N] 2+ -core with a bidentate dithiocarbamate (DTC) and tridentate PNP-type bisphosphine chelates [30]. One example is the 99m Tc-nitrido complex DBODC5 ( 99m Tc-[bis (dimethoxypropylphosphinoethyl)-methoxyethylamine (PNP5)]-[bis (N-ethoxyethyl)-dithiocarbamate (DBODC)] nitride (N-PNP5-DBODC or N-DBODC5) (Table 2), which demonstrated high cardiac uptake and retention for more than 2 hours and nearly complete elimination of hepatocyte activity by two hours in Sprague-Dawley (SD) rats [31, 32].…”
