Chemical Studies of Formosan Cobra (Naja naja atra) Venom. Part I. Chromatographic Separation of Crude Venom on CM‐Sepadex and Preliminary Characterization of its Components

Abstract: The crude venom of Formosan cobra (Naja naja atra) was separated into 12 fractions by CM‐Sephadex (C‐50) chromatography with ammonium acetate buffer by two‐stage gradient elution. Fr. I was proved to be a mixture of nucleotides and nucleosides. The toxicity was found in Fr. VIII (neurotoxic), X (cardiotoxic), XI (cardiotoxic), and XII (cardiotoxic). Glycerophosphatase, alkaline phosphomonoesterase, and 5′‐nucleotidase were found in Fr. III, V and IX, respectively. Phosphodiesterase was distributed in Frs. VI a… Show more

“…Phosphodiesterase activity was determined by a method modified from Lo et al (29). One hundred microliters of sample was added to an assay mixture containing 0.5 mL of 2.5 mM calcium bis-p-nitrophenylphosphate, 0.3 mL of 0.01 M magnesium sulfate and 0.5 mL of 0.17 M veronal buffer, pH 9.0.…”

“…Phosphodiesterase activity was determined using disodium-p-nitrophenol phosphate as substrate (Lo et al, 1966). Reaction mixture containing 0.5 mL of 0.25% venom solution/fraction was added to an assay mixture containing 0.5 mL 0.0025 M sodium-p-nitrophenyl phosphate, 0.3 mL 0.01 M MgSO4, 0.5 mL 0.17 M Tris-HCl buffer (pH 8.0).…”

“…Cats, being relatively resibcant to cobra reurotoxin, die of cirulatory failure before the neurotoxin induces peripheral respiratory paralysis (Lee and Peng, 1961;Lee, 1963). Several active components such as neurotoxin, cardilotoxin, phospholipase A, and some proteins having other enzymatic a-Alvfties have been separated from cobra venom (Lo et al, 1966; also see Meldrum, 1965;Boquet, 1966). However, it has not been established as to which component (s) or to what extent these components are responsible for the cardiovascular effects caused by cobra venom.…”
