Keratin is a fibrous, structured protein that constitutes the basis of different structures such as nails, hair, beak, wool, feathers, claws, and horns. Until now, keratin isolation has been carried out from waste wool, feather, hoof and hair. However, the development of effective techniques to obtain keratin without any damage to the secondary structure of the protein remains a challenging task. Here in, two distinct keratin isolation methods (Na2S and EDTA) were compared for the first time from Domestic Goose (Anser domesticus) feathers. Kjeldahl method was used for the determination of crude protein from the obtained keratin powders by two methods, and antioxidant activity of keratin powders was conducted. Our findings suggest that Na2S method displayed the best keratin yield (86.34%) from goose feather; however, antioxidant activity of EDTA method was nearly three times higher than Na2S method, which could be taking into consideration as a potential keratin source for future studies.

In silico Prediction and In vitro Antioxidant Activities of Two Jujube Fruits from Different Regions

Jujube is a fruit rich in antioxidant compounds and vitamin C. In this way, it can prevent cell damage by fighting free radicals. In the study, antioxidant activities, total phenolic and total flavonoid compound amounts of methanolic extracts of jujube fruits were determined. In addition, their biochemical compositions were determined using HPLC. Also, an in silico prediction study of the identified active ingredients was performed to evaluate antioxidant, antiradical, antibacterial, antifungal, anti-inflammatory, antimutagenic, and membrane integrity antagonist properties. For the determination of antioxidant capacity, ferric reducing power (FRAP) and cupric ion reducing capacity (CUPRAC), DPPH radical scavenging activity and β-carotene-linoleic acid assay were used. The amounts of total phenolic and the total flavonoid compounds of the extracts were determined as gallic acid equivalent (GAE) and as quercetin equivalent (QE). The methanolic extract of jujube collected from Antalya contains 580 μg GAE.mg-1 and 240 μg QE.mg-1, and the methanolic extract of jujube collected from Denizli contains 900 μg GA.mg-1 and 380 μg QE.mg-1. The IC50 values of Antalya and Denizli methanolic extracts according to the DPPH scavenging assay were 10.34 and 9.82, respectively. Gallic acid, catechin, caffeic acid, coumaric acid, ferulic acid and cinnamic acid were detected by HPLC. In addition, the in silico effects of these molecules were estimated with the PASS online prediction program. As a result, it was seen that Denizli jujube had a higher antioxidant effect than Antalya jujube.

The genus Nepeta belongs to the largest Lamiaceae family, with 300 species, which are distributed throughout the various regions of Africa, Asia, India, and America. Along with other plant families distinguished by their medicinal and therapeutic values, the Nepeta genus of Lameaceae remains relatively valuable. Hence, the phytochemicals of N. paulsenii Briq. were extracted using different plant parts, i.e., leaves, stem, roots, flowers, and the whole plant by using various solvents (ethanol, water, and ethyl acetate), obtaining 15 fractions. Each extract of dried plant material was analyzed by FT-IR and GC-MS to identify the chemical constituents. The cytotoxicity of each fraction was analyzed by MTT assay and mitochondrial membrane potential and nuclear condensation assays against lung cancer cells. Among the ethyl acetate and ethanolic extracts, the flowers showed the best results, with IC50 values of 51.57 μg/mL and 50.58 μg/mL, respectively. In contrast, among the water extracts of the various plant segments, the stem showed the best results, with an IC50 value of 123.80 μg/mL. 5-flourouracil was used as the standard drug, providing an IC50 value of 83.62 μg/mL. The Hoechst 33342 stain results indicated apoptotic features, i.e., chromatin dissolution and broken down, fragmented, and crescent-shaped nuclei. The ethanolic extracts of the flowers showed more pronounced apoptotic effects on the cells. The mitochondrial membrane potential indicated that rhodamine 123 fluorescence signals suppressed mitochondrial potential due to the treatment with the extracts. Again, the apoptotic index of the ethanolic extract of the flowers remained the highest. Hence it can be concluded that the flower part of N. paulsenii Briq. was found to be the most active against the A459 human lung cancer cell line.
