Nyamplung (Calophyllum inophyllum L) adalah tumbuhan yang tersebar luas di Indonesia, terutama ditemukan di daerah pantai berpasir. Tumbuhan dengan pohon tinggi sedang ini sudah lama digunakan masyarakat sebagai obat. Keragaman metabolit sekunder yang dikandungnya memiliki potensi untuk dikembangkan sebagai bahan baku obat untuk industri obat. Artikel ini ditulis berdasarkan literatur ilmiah untuk menjelaskan hubungan pemanfaatan Nyamplung (Chalohyllum inophyllum L.) dengan bioaktifitasnya sehingga pemanfaatannya sebagai obat dapat dikembangkan. Berdasarkan sejumlah penelitian tentang metabolit sekunder ditemukan senyawa terutama kelompok xanthone, kumarin, triterpenoid dan flavonoid. Senyawa ini memiliki bioaktivitas termasuk antikanker, antivirus, anti-HIV, antibakteri, antijamur, dan antiinflamasi.

Introduction: Xanthones are metabolites with a variety of biological properties. The Clusiaceae family, which until recently included the genus Calophyllum, is recognised for its production of monohydroxylated and polyhydroxylated xanthones. Presently, C. brasiliense is the only Calophyllum spp. known to occur in the Yucatan peninsula. Objective: To use a combination of traditional phytochemical methods and carbon-13 nuclear magnetic resonance ( 13 C-NMR) dereplication analysis to identify xanthones in the stem bark of C. brasiliense. Material and methods: Initial fractionation and purification of the stem bark extract of C. brasiliense produced macluraxanthone (1). Additional xanthones, together with chromanones and terpenoids, were identified using 13 C-NMR dereplication analysis in different semipurified fractions obtained from the low and medium polarity fractions of the stem bark extract of C. brasiliense.Results: Initial identification of macluraxanthone (1) was confirmed by 13 C-NMR dereplication analysis; additionally, 13 C-NMR dereplication analysis allowed the identification of a number of monohydroxylated and polyhydroxylated xanthones, together with chromanones and terpenoids. Conclusion:This study confirms C. brasiliense as a rich source of xanthones and the 13 C-NMR dereplication analysis as a suitable method to quickly identify the presence of different families of secondary metabolites in semipurified fractions.

Anti-proliferative metabolites from the root of calophyllum inophyllum L.

This study aimed to isolate secondary metabolites from the root of C. inophyllum L. and to evaluate their anti-proliferative activities against hepatocellular carcinoma and colorectal cancer cells. Chromatographic fractionation of the n-hexane and ethyl acetate crude extracts led to the isolation of two new (1 and 2) and twelve known xanthones (3-14). In addition, metabolites 4, 6 and 8 were isolated from the genus Calophyllum for the first time. All metabolites were evaluated for their anti-proliferative activity against Hep-G2 and HCT-116 cell lines. Metabolites 5 and 14 showed the most active metabolites against both cancer cells, as well as they displayed strong activities toward HCT-116 with the same IC50 value of 3.04 µM. Moreover, both of them exhibited potent activities against Hep-G2 with IC50 values of 6.07 and 5.94 µM, respectively. Further, metabolite 14 was subjected to western blot analysis to study its mechanism of action against Hep-G2 cells. The result demonstrated that 14 induced apoptosis in a concentration-dependent manner by the down-regulation of the expression of both Bcl-2 and Bcl-XL proteins.
