The study highlights for the first time the potential role of Caryota mitis and Caryota urens (family Arecaceae) in cancer chemoprevention as NQO1 enzyme inducers. Among the tested leaf extracts of different solvent polarities, C. urens petroleum ether leaf extract showed the most potent induction of NQO1 enzyme activity (4.79 times to vehicle control) via DCPIP assay and a significant difference was observed at concentration 25 μg/mL via NQO1 western blot analysis. First comparative GC/MS analysis of both species' petroleum ether leaf extracts was conducted to reveal for 35 vs 34 compounds in C. mitis and C. urens unsaponifiable matter, respectively, and 29 vs 15 fatty acids as methyl esters, respectively. Isolation attempts from the active C. urens extract resulted in the identification of five compounds i.e. oleanolic acid (T1), ursolic acid (T2), α-tocopherol (T3), palmitic and stearic acids (T4 & T5) using different spectroscopic techniques; where T1 and T3 are first time to be identified in C. urens. Assessment of the isolated compounds' chemopreventive activity was conducted via in silico molecular modeling against KEAP-1 -Nrf2 complex involved in of NQO1 where T1, T2 and T3 showed promising docking score of -7.7, -7.9 and -7.05 kcal/mol, respectively, compared to -8.2 kcal/mol for 4'-bromoflavone as a positive control.

Background: Caryota urens flowers and Hyophorbe lagenicaulis leaves have huge beneficial properties, medicinal uses and rich phyto-pharmacological constituents. Nutraceutical potentiality of the selected plants on the basis of phytochemicals present in the aerial parts of Caryota urens and Hyophorbe lagenicaulis were taken as the background of the study. Aim: The present study investigates the nephroprotective potentiality of Caryota urens flowers and Hyophorbe lagenicaulis leaves against cisplatin and gentamycin induced nephrotoxicity. Materials and Methods:The extract of Caryota urens flowers and Hyophorbe lagenicaulis leaves were screened for preliminary phytochemical analysis. The in vitro antioxidant activity of Caryota urens flower extract (CUFE) and Hyophorbe lagenicaulis leaves extract (HLLE) was performed by DPPH and H 2 O 2 radical scavenging activity. In vivo acute oral toxicity test was performed by following OECD guideline 420. The screening models for cisplatin and gentamycin induced nephrotoxicity by using albino Wistar rats were also carried out. Results: In in vitro antioxidant studies CUFE and HLLE (*p<0.01 and **p<0.001) for both standard and test drugs. It also act as potent nutraceuticals in cisplatin and gentamycin induced nephrotoxicity (**p<0.001). No major side effects were seen in any of the experimental animal during acute oral toxicity. Conclusion: CUFE and HLLE (1:1) have nutritionally rich source of dietary food supplements and due to this they are also helpful in managing the commercial drugs induced nephrotoxicity.

Caryota mitis and Caryota urens (F. Arecaceae) are well reported in Ayurvedic medicine and involved in several edible food products. Herein, the first comparative profiling of their leaf and fruit metabolites of both species via HR-UPLC/PDA/ESI-MS and multivariate data analyses is presented. A total of 142 metabolites were detected with seven reported for the first time in F. Arecaceae and a novel O-caffeoylshikimic acid conjugate. Screening of plants' leaf crude extracts via in vitro DCPIP kinetic assay revealed the induction of phase II cytoprotective enzyme NQO1 by 4.5-to 5-fold versus control, suggestive of potential chemopreventive activity. Two novel sulfated flavonols that is quercetin-3-O-sulfate-4′-O-rhamnosyl (1→6)βd-glucoside (F1) and kaempferol-3-O-sulfate-4′-O-rhamnosyl(1→6)βd-glucoside (F3) in addition to another five known flavonoids were isolated from C. mitis ethanol extract and identified via MS and NMR spectroscopic techniques. Among isolated compounds, F1 and F3 exhibited the highest docking score as KEAP-1 inhibitors and Nrf2 activators posing them as potential chemopreventive drug leads. Practical applicationsThe study extends the usages of this edible less explored Caryota species to a potential cancer chemopreventive action. Guided by the extensive chemical information presented herein, additional uses could be suggested for these plants with 142 identified metabolites including androst-en-diol that has aphrodisiac and muscle building effects. The presented multivariate data analyses could aid phytochemists in plants classification and mapping (chemotaxonomy) since several metabolites are reported herein for the first time either in family or genus.
