Chemical Constituents from Sambucus adnata and Their Protein-Tyrosine Phosphatase 1B Inhibitory Activities

Abstract: Type-2 diabetes mellitus (T2DM) is a chronic metabolic disorder characterized by b-cell dysfunction and insulin resistance, and its prevalence has reached epidemic proportions worldwide.1-3) Protein-tyrosine phosphatase 1B (PTP1B) is a major non-transmembrane phosphotyrosine phosphatase expressing ubiquitously in the classical insulin-targeted tissues 4,5) and functions as a negative regulator of the insulinsignaling pathway.6,7) Inhibition of PTP1B has been demonstrated to be an effective therapeutic approach… Show more

“…The nonenzymatic hydrolysis of 2 mM p -NPP was corrected by measuring the increase in absorbance at 405 nm, which was obtained in the absence of PTP1B. In this study, ursolic acid, a known PTP1B inhibitor, was used as the positive control and exhibited an IC 50 (50% inhibition concentration) of 4.3 μM, which was consistent with a previous report [16]. …”

“…These data were indicative of the presence of an isovaleryl moiety. The signals at 22.3 (2C), 25.6, 42.9, and 171.7 in the 13 C NMR spectrum supported this assumption. 14 Moreover, the presence of 2 acetoxy groups was evident from the signals at δ H 2.05 (3H) and 2.04 (3H) and the corresponding carbon resonances at δ C 21.0, 20.8, and 170.2 (2C).…”

“…Boehmenan, isolated from Hibiscus taiwanensis (Malvaceae), was moderately cytotoxic to the A549 (human lung adenocarcinoma epithelial cell line) and MCF-7 cell lines with EC 50 values of 18.4 and 10.9 μg/mL, respectively, and inhibited HIV replication in H9 lymphocyte cells with an IC 50 value of 19.43 μg/mL [66]. Moreover, boehmenan obtained from a methanolic extract of whole plants of Sambucus adnata (Caprifoliaceae) inhibited PTP1B (protein-tyrosine phosphatase 1B) with an IC 50 value of 43.5 g/mL [67]. Erythro-carolignan E (13) showed cytotoxic activity against A549 and MCF-7 cell lines with EC 50 values of 10.6 and 8.5 g/mL, respectively [66].…”
