Codon usage bias influences the genetic features prevalent in genomes of all the organisms. It also plays a crucial role in establishing the host-pathogen relationship. The present study elucidates the role of codon usage pattern regarding the predilection of fungal pathogens Aspergillus flavus, Aspergillus niger, Fusarium oxysporum and Colletotrichum gloeosporioides towards host plant Zingiber officinale. We found a similar trend of codon usage pattern operative in plant and fungal pathogens. This concurrence might be attributed for the colonization of fungal pathogens in Z. officinale. The transcriptome of both plant and pathogens showed bias towards GC-ending codons. Natural selection and mutational pressure seem to be accountable for shaping the codon usage pattern of host and pathogen. We also identified some distinctive preferred codons in A. flavus, F. oxysporum and Z. officinale that could be regarded as signature codons for the identification of these organisms. Knowledge of favored, avoided and unique codons will help to devise strategies for reducing spice losses due to fungal pathogens.

This study evaluates the volatile metabolic constituents and anticancer potential of essential oils distilled from the rhizomes of four Malaysian Zingiber officinale Roscoe (Zingiberaceae family) varieties (Bentong (BE), Cameron Highlands (CH), Sabah (SA), and Bara (BA)). The ginger essential oils were analyzed by gas chromatography coupled with quadrupole mass spectrometry (GC qMS). A total of 58 secondary compounds were tentatively identified, representing 82.6–87.4% of the total ion count. These metabolites comprise mainly of monoterpene hydrocarbons (19.7–25.5%), oxygenated monoterpenes (23.6–33.7%), sesquiterpene hydrocarbons (21.3–35.6%), oxygenated sesquiterpenes (1.5–3.9%), and other minor classes of compounds (0.7–2.7%). Principal component analysis (PCA) enabled differentiation of the analyzed ginger essential oils according to their varieties, with respect to their metabolites and relative quantities. The antiproliferative activity against the HeLa cervical cancer cell line was investigated via the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The oils were found to exhibit strong antiproliferative activities with IC50 values of 23.8, 35.3, 41.3, and 42.5 μg/mL for BA, BE, SA, and CH, respectively. These findings suggest that the differences among the secondary metabolites and their abundance in different varieties of Z. officinale essential oils appear to be related to their antiproliferative potential. The strong antiproliferative effects of these oils signified their potential in the prevention and chemotherapy of cervical carcinoma treatment.

The constant increase in the emergence of multidrug resistance among Staphylococcus aureus strains threatens public health. The study aimed at evaluating the anti-S. aureus activity, pharmacokinetic properties and interactions of compounds from Erianthemum dregei with proteins in S. aureus. Anti-S. aureus activity was investigated by broth dilution method while gas chromatography-mass spectrometry (GC-MS) was used to identify the compounds. The drug-likeness, pharmacokinetic and toxicity profiles of the compounds were predicted by SwissADME and PreADMET tools. AutoDock Vina was used to assessing the binding affinities of the docked ligand-receptor complexes. The extract revealed the minimum inhibitory concentration value of 0.78 mg/mL. Phytol (93.58%) and 3-tetradecyn-1-ol (6.42%) were the revealed constituents. In-silico predictions suggested both compounds to have drug-like properties as they adhered to the Lipinski's rule of five. Phytol was found to have non-mutagenic effects, while 3-tetradecyn-1-ol was predicted to be mutagenic. The compounds were non-carcinogenic on mice model and carcinogenic on rat`s. Phytol has a binding affinity to DNA-gyrase and FtsZ with docking energy values of -4.1 and -5.3 kcal/mol, respectively, whereas the docking scores for 3-tetradecyn-1-ol against DNA-gyrase and FtsZ were -3.9 and -5.0 kcal/mol. The results revealed the extract to have a noteworthy activity against S. aureus, with its identified compounds having desirable pharmacokinetics. INTRODUCTION:Staphylococcus aureus is a member of the family Micrococcaceae that are characterized as catalase-positive and Grampositive aerobic cocci 1 . S. aureus is known known to cause clinical manifestations such pneumonia, endo-carditis, toxic shock syndrome, cellulitis, abscesses and impetigo at varying severity in humans 2 . Its infections are most predominant in developing countries, especially among the elderly, young children, and immunosuppressed people 3 .
