Plant volatile sesquiterpenes (PVSs) play important roles in chemical plant defense. However, it is difficult to isolate sufficient PVSs for deep investigations due to their low contents and chemical and physical properties close to those of other lipids. The extracts of Stellera chamaejasme L. exhibit insecticidal, fungicidal, and allelopathic activities. In this study, we identified three sesquiterpene synthase genes (SchTPS5, SchTPS6, and SchTPS7) from S. chamaejasme L. SchTPS7 is an α-farnesene synthase. SchTPS5 and SchTPS6 are two catalytically promiscuous sesquiterpene synthases, and α-cadinol and τ-muurolol are the predominant products for both of them in Saccharomyces cerevisiae. This study, for the first time, reports plant sesquiterpene synthases capable of producing α-cadinol and/or τ-muurolol in a heterologous host. More intriguingly, seven out of eight products of SchTPS6 in S. cerevisiae possess various insecticidal, fungicidal, and herbicidal activities. Building on this finding, we used SchTPS6 to construct an engineered S. cerevisiae for the production of these sesquiterpenes. The titers of two major products α-cadinol and τ-muurolol, respectively, reached 46.2 ± 4.0 and 11.2 ± 1.4 mg/L in a flask. This study lays a foundation for the development of new agrochemical mixtures.

Black pepper (Piper nigrum) is a spice commonly used in kitchens throughout the world. Black pepper production is devastated by a range of pathogenic agents, including Phytophthora capsici and Meloidogyne incognita. Many efforts have been directed towards finding black pepper cultivars that are resistant to these pathogens. In this work, a 39‐accession germplasm panel of species in the Piper family collected throughout Vietnam was described. Preliminary tests using P. capsici inoculation onto leaves were carried out to identify potentially resistant accessions. Next, candidate plants were inoculated with P. capsici mycelial suspension and survival rates were assessed 15, 30 and 45 days postinoculation. In addition, Piper plants were challenged with M. incognita by adding larvae/juveniles to growing pots. Resistance to M. incognita was determined by the number of root galls and the percentage of plants with yellow leaves 1, 2 and 4 months after treatment. Piper accessions were also subjected to a 4‐day waterlogged treatment. Two accessions (HUIB_PH30 and HUIB_PD36) demonstrated high levels of resistance to all biological and water stresses. Micromorphological characterizations revealed that the amount of intercellular spaces in the root cortex correlated with the resistance to P. capsici and waterlogging tolerance. Hence, the abundance of intercellular spaces can serve as a guide for further selection of black pepper accessions that are resistant to common diseases and tolerant to waterlogged conditions.

In this work, essential oils extracted from roots and aerial parts of Inula graveolens by hydrodistillation and their fractions obtained by chromatographic simplification were first investigated for their chemical composition by GC/MS and then evaluated for the first time for their repellency and contact toxicity properties against Tribolium castaneumadults. Twenty-eight compounds were identified in roots essential oil (REO), which accounted for 97.9 % of the total oil composition, with modhephen-8-β-ol (24.7 %), cis-arteannuic alcohol (14.8 %), neryl isovalerate (10.6 %) and thymol isobutyrate (8.5 %) as major constituents. Twenty-two compounds were found in the essential oil from aerial parts (APEO), which accounted for 93.9 % of the total oil, with borneol (28.8 %), caryophylla-4(14),8(15)-dien-6-ol (11.5 %), caryophyllene oxide (10.9 %), τ-cadinol (10.5 %) and bornyl acetate (9.4 %) as main compounds.REO and APEO displayed stronger repellency after 2 h of exposure (80.0 and 90.0 %, respectively) against T. castaneum at the concentration of 0.12 μL/cm 2 . After fractionation, fractions R 4 and R 5 exhibited greater effects (83.3 % and 93.3 %, respectively) than the roots essential oil. Furthermore, the fractions AP 2 and AP 3 showed higher repellency (93.3 and 96.6 %, respectively) than the aerial parts oil. The LD 50 values of oils from roots and aerial parts topically applied were 7.44 % and 4.88 %, respectively. Results from contact toxicity assay showed that fraction R 4 was more effective than the roots oil with LD 50 value of 6.65 %. These results suggests that essential oils of roots and aerial parts from I. graveolens may be explored as potential natural repellent and contact insecticides against T. castaneum in stored products.
