Chemerin Effect on the Endometrial Proteome of the Domestic Pig during Implantation Obtained by LC-MS/MS Analysis

Abstract: Chemerin (CHEM) is a hormone mainly expressed in adipocytes involved in the regulation of energy homeostasis and inflammatory response. CHEM expression has been demonstrated in the structures of the porcine hypothalamic-pituitary-gonadal axis, as well as in the uterus, trophoblasts and conceptuses of pigs. In this study, we performed high-throughput proteomic analyses (liquid chromatography with tandem mass spectrometry, LC-MS/MS) to examine the influence of CHEM (400 ng/mL) on differentially regulated protein… Show more

“…We hypothesized that the energy supply was necessary after the initiation of implantation. In addition to the change in the endometrial signaling pathway, there was also an influence of the number of conceptuses [ 37 ]. Variations within the number of conceptuses result in variable levels of their signals in the uterine lumen; therefore, the maternal response could be different, which is also related to the number of corpora lutea.…”

“…Variations within the number of conceptuses result in variable levels of their signals in the uterine lumen; therefore, the maternal response could be different, which is also related to the number of corpora lutea. Variation in the number of corpora lutea may result in different levels of P4 affecting the endometrium [ 37 ].…”

“…Proteomic analysis was performed at the Mass Spectrometry Laboratory at the Institute of Biochemistry and Biophysics of the Polish Academy of Sciences. Samples were digested using the filter-aided sample preparation (FASP) method 31 according to the previously described protocol 32 , with minor modifications. In brief, 150 µg of total proteins was incubated for an hour with 20 mM tris(2-carboxyethyl)phosphine (TCEP) at 60 °C to reduce cysteine residues.…”

“…TMT-labeled peptides were fractionated using the high-pH reversed-phase method into 25 fractions. The system and gradient parameters are described in our previous publication [28].…”
