Chartarlactams Q−T, Dimeric Phenylspirodrimanes with Antibacterial and Antiviral Activities

Abstract: Four new phenylspirodrimane‐type dimers, namely chartarlactams Q−T, along with stachyin B were isolated from the fermentation broth of a sponge‐derived fungus Stachybotrys chartarum WGC‐25 C‐6. Chartarlactams Q−T were structurally featured by the dimerization of two units of phenylspirodrimane linked by a C−N bond. Their structures were determined on the basis of extensive spectroscopic analysis, while quantum ECD calculation and modified Mosher's method were used for the assignment of absolute configurations.… Show more

“…Some of these compounds exhibited antihyperlipidemic effects on HepG2 cells, such as inhibition of intracellular triglyceride (TG) levels and reduction of total cholesterol (TC), but the mechanism of antihyperlipidemic activity remains unknown. The same group reported four new phenylspirodrimane-type dimers in 2020, which showed antibacterial (MIC: 4−16 μg/mL) and antiviral activities (Liu et al 2020).…”

“…Four new phenylspirodrimane-type dimers namely chartarlactams Q–T ( 189 – 192 ), were isolated from the fermentation-broth of a sponge-derived fungus Stachybotrys chartarum WGC-25C-6. Compounds 189 – 191 showed moderate inhibition against bacterial pathogen Staphylococcus aureus with MIC values ranging from 4 to 16 μg/mL, while compound 192 exhibited significant inhibition toward ZIKV virus [ 53 ]. Meanwhile, chartarene D ( 193 ), was isolated from the sponge ( Niphates recondite )-associated fungus Stachybotrys chartarum [ 12 ].…”

“…Envelope (E) protein in ZIKV virus has a remarkable role in membrane fusion and binding to target the host cells receptors, whereas NS5 protein a ZIKV genome encoding non-structural proteins that serves as template for the (+)ssRNA genomic RNA synthesis and viral polyprotein synthesis. Hence, this metabolite prohibited ZIKV virus by multi-targets to block RNA replication and viral entrance [ 86 ]. Biosynthetically, they are generated from 3 that undergoes nucleophilic cyclization to give I (hemiacetal intermediate).…”
