Characterizing a rat Brca2 knockout model

Abstract: Evidence exists that BRCA2 carriers may have an elevated risk of breast, ovarian, colon, prostate, and pancreatic cancer. In general, carriers are defined as individuals with protein truncating mutations within the BRCA2 gene. Many Brca2 knockout lines have been produced and characterized in the mouse. We previously produced a rat Brca2 knockout strain in which there is a nonsense mutation in exon 11 between BRC repeats 2 and 3, and a truncated protein is produced. Interestingly, while such a mutation in homoz… Show more

“…It is intriguing that zep display renal failure and is embryonic lethal, features that are distinct from brca2 Q658X and brca2 ZM_00057434 . Across vertebrate models to date, BRCA2 null alleles are embryonic lethal in mouse and humans while viable in rats (Moynahan, 2002; Cotroneo, et al, 2007). Identification of whether zep represents a genetic null, hypomorphic, or hypermorphic allele will require many additional studies, and these will be necessary to further elucidate the molecular consequences that lead to the zep phenotype.…”

“…Full length Brca2 protein was identified from testicular extracts from Brca2 +/+ and Brca2 +/-rats, and was absent in Brca2 -/-testes, providing validation of this model and a unique opportunity to assess Brca2-associated breast cancer susceptibility [52].…”

“…Phenotypic evaluation of Brca2 -/-rats having the SD genetic background revealed several abnormalities [52]. Compared with Brca2 +/+ and Brca2 +/-littermates, Brca2 -/-rats were significantly smaller and were infertile, as a consequence of impaired gonad Gould, Michael N. W81XWH-04-1-0436 development.…”

“…Animals have a substantial reduction in lifespan that could be due, at least in part, to an increase in cancer development in multiple organs (stomach, bone, ovary, etc.). Tumors arising most frequently were osteosarcomas and ovarian tumors [10]. The exhaustive genetic analysis of breast cancer patients, which allow us to identify novel mutations, together with the use of animal models is a two-way road.…”
