Characterization of vine varieties by SSR markers

Abstract: Nowadays, there are well known PCR-based techniques to detect polymorphism in plants. Marker technologies which are applied in breeding and varietal characterization can be exchanged across laboratories with standardization to yield reproducible results. This article describes applying of simple sequence repeats (SSR) markers in European laboratories, in which the reproducibility of these popular markers are examined. One of the European Union interests is characterization of grape varieties and preparation of… Show more

“…Using nuclear microsatellite loci (nSSR) [ 18 , 19 , 20 ] and chloroplast (cpSSR) [ 21 , 22 , 23 ] DNA which were intended for genetic identification of grapes variety is one of the approaches to DNA authentication of wines [ 1 , 2 , 3 , 4 , 5 , 6 , 8 , 9 , 10 , 11 , 12 , 13 , 17 , 24 , 25 , 26 ], a sequential procedure which is carried out with separately taken SSR-marker primer set, and not multiplex PCR.…”

“…The genome of Vitis vinifera L. contains a huge number of polymorphic microsatellite loci, the allelic diversity of which can be used to expose the genetic structure of grape germplasm collections maintained ex situ, as well as for detection of doublets and varietal identification (Dokupilová et al, 2013).…”

“…Геном V. vinifera содержит множество микросателлитных локусов, в высшей степени полиморфных (Dokupilová et al, 2013). Исследованиями Thomas et al (1993а) было показано, что в геноме винограда преобладают микросателлитные локусы, содержащие дву-и трехнуклеотидные повторы (GT)n, (GA), (CAC), (GACA)n, (GATA)n. При этом установлено, что повторы GA and GT наиболее многочисленны и распространены по всему геному.…”
