Characterization of Two F‐Actin‐Binding and Oligornerization Sites in the Cell‐Contact Protein Vinculin

Abstract: Vinculin, a structural protein of animal cells, is critically involved in the assembly of microfilament/ plasma membrane junctions at cell contacts. To understand its role in organizing the distal portions of microfilaments into specific, morphologically distinct structures at these sites in more detail, we characterized its interaction with filamentous actin and with itself by means of in vitro assays. Using recombinant proteins comprising different parts of the vinculin tail fused to the maltose-binding prot… Show more

“…After 16 h cells were treated with the membrane-permeant cross-linker dithiobis(succinimidyl propionate) (DSP; Pierce) essentially as described in Huttelmaier et al (37). In brief, cells were incubated with DSP for 30 min.…”

“…For this assay (Fig. 5B), we used full-length metavinculin and vinculin and probed their binding to actin filaments as an indicator of the open conformation, with the actin binding site in the tail (11,12,20,21) accessible. Cosedimentation with F-actin was performed in the presence of increasing amounts of the talin peptide.…”

“…It is a multiligand protein, and binding of VH to talin or ␣-catenin targets vinculin to microfilament-associated cell adhesions (7)(8)(9)(10)). Anchorage of microfilaments is mediated by VT that harbors two actin binding regions (11,12). Through the recruitment of the vasodilator-stimulated phosphoprotein VASP and the Arp 2/3 complex to the hinge region, vinculin is also tightly linked to actin dynamics at cell adhesion sites (13)(14)(15).…”

“…Each discrete vinculin domain recognizes multiple binding partners: the vinculin head (Vh) 4 binds to talin, ␣-actinin, and IpaA (14 -16); the prolinerich linker interacts with vasodilator stimulated phosphoprotein (VASP), CAP/ponsin, vinexin, and the Arp2/3 complex (17)(18)(19)(20); the vinculin tail (Vt) associates with filamentous actin (F-actin), phosphatidylinositol (4,5)-bisphosphate, and paxillin (21)(22)(23). In the autoinhibitory form of vinculin, tight intramolecular interactions between Vh and Vt occlude binding to most of its ligands (13).…”
