Characterization of Tumor Suppressive Function of cornulin in Esophageal Squamous Cell Carcinoma

Abstract: By using cDNA microarray analysis, we identified cornulin (CRNN) gene was frequently downregulated in esophageal squamous cell carcinoma (ESCC). In the present study, we investigated the role of CRNN in ESCC development. The results showed that CRNN was frequently downregulated in primary ESCCs in both mRNA level (26/56, 46.4%) and protein level (137/249, 55%), which was significantly associated with lymph node metastases (P=0.027), advanced clinical stage (P=0.039), and overall survival rate (P<0.001). “…To resolve the question if the CRNN mRNA reduction in AD skin is caused by a gene polymorphism or whether other factors are involved, as is the case with FLG , whose expression is diminished by Th2 cytokines [1,17] , further studies are necessary. The reduction of CRNN mRNA and protein levels was previously reported in primary esophageal squamous cell carcinoma (SCC) and indicat-ed as an independent prognostic factor for esophageal SCC [36] . Furthermore, in laryngeal SCC, the CRNN transcript is downregulated [37] .…”

“…Furthermore, in laryngeal SCC, the CRNN transcript is downregulated [37] . It has been demonstrated that CRNN is a tumor-suppressor gene [36] . On the other hand, there are studies reporting an increased risk of SCC among AD patients [38,39] .…”

“…In a recent study by Chen et al it was shown that cornulin is a potential tumor suppressor and it regulates cell cycle progression at G1/S checkpoint by upregulating P21 WAF1/CIP1 and Rb. Thus it exerts its tumor suppressor effect by inhibiting G1 to S phase transition in cell cycle [ 44 ].…”

“…37 Cornulin is involved in cell cycle arrest at the G1/S checkpoint by upregulating the expression of p21(WAF/CIP). 38 Progression of renal failure in a subtotal nephrectomy model in p21 knockout mice was slower and associated with a 5-fold increase in epithelial surface compared with wild-type mice, suggesting that epithelial repair is hampered in the wild-type strain. Epithelial cell cycle arrest has been identified as a major promotor of kidney fibrosis.…”
