The maintenance of the recombinant plasmid p I J 2 in chemostat cultures of Streptomyces lividans 66 (~1 5 2 ) was investigated. The presence of tho plasmid coding for a ncomycin phosphotransferase was detected by plating samples from the continuous cultures on nonselective and selective agar medium containing neomycin.The plasmid was lost from the host strain under all conditions tested. However, the kinetics of the plasmid segregation from the chemostat populations were dependent on the growth-limiting substrate of the medium, the dilution rate of the continuous culture and the cultivation ternperature.Size differences between the original plasmid and plasmid DNA isolated from neomycinresistant clones after long-term chemostat cultivation were not observed. In neomycin-sensitive clones no extrachromosomal DNA was found.Recently a substantial literature has accumulated concerning the structural stability and the maintenance of plasmids in their host organisms. Continuous culture techniques proved as a suitable tool in studying these problems (for review DYKHUIZEN and HARTL 1983). Knowledge about stability and maintenance is relevant particularly in the case of vector and recombinant plasmids. For the successful use of a plasmid-containing bacterial strain in large-scale fermentation of a plasmid-coded product it is necessary that the recombinant plasmid is both structurally stablc and maintained at a sufficient copy number.It was, for example, impossible to produce tryptophan in a continuous fermentation

Bacteriophage O2 multiplies normally on Oerskovia turbata IMET 47 153. It has a burst size of about 100 p.f.u. per infected cell and a latent period of 100 min at 30 degrees C. On Oerskovia xanthineolytica IMET 47 383 clear spots were formed after addition of high phage concentrations onto agar top layers. By phase contrast observation, and measurement of the optical density of infected cultures, it was found that the clearing effect on strain IMET 47 383 was due to lysis-from-without. Phage O2 adsorbs and injects its DNA into cells of strain IMET 47 383 but phage multiplication does not occur, and the phage DNA becomes degraded. Inhibition of phage DNA injection by the combined action of xanthotoxin -- u.v. irradiation abolished the clearing activity of phage lysates. Therefore, both adsorption and DNA injection seem to be prerequisites for the release of a lytic activity out of the phage particle, which is responsible for the clearing effect on strain IMET 47 383.
