Characterization of the Salmonella typhimurium pagC/pagD chromosomal region

Abstract: The PhoP/PhoQ two-component system regulates Salmonella typhimurium genes that are essential to bacterial virulence and survival within macrophages. The best characterized of these PhoP-activated genes (pag) is pagC, which encodes a 188-amino-acid envelope protein (W. S. Pulkkinen and S. I. Miller, J. Bacteriol. 173:86-93, 1991). We here report the identification of four genes (pagD, envE, msgA, and envF) located 5 to pagC. Each gene is transcribed from its own promoter, two of which (msgA and pagD) were defin… Show more

“…The spaN and sipB genes were shown to have a role in entry into non-phagocytic cells and killing of macrophages [37]. Additionally, there are genes required for the survival within macrophages, including pagC [6], msgA [38], and spiA [39]. Others have a role in iron acquisition, including sitC [40] and iroN [41].…”

“…Fourteen genes-invA, orgA, prgH, spaN (invJ ), tolC, sipB, pagC, msgA, spiA, sopB, lpfC, pefA, spvB, and sifA-assayed in this study encode products that are associated with cellular invasion, survival within a cell, and adhesin or pili production (2, 3,4,5,7,8,12,16,18,19,26,27,29). The invA, prgH, spaN (invJ ), sipB, spiA, and sifA genes are also associated with type III secretion systems (TTSSs) (18,28,30), complex structures of more than 20 proteins that are used in protein delivery (10).…”

“…Other targeted genes encoded other traits thought to be important to virulence, including sitC (11) and iroN (1), both involved in iron acquisition, and cdtB, a putative toxin-encoding gene (9). All of the genes except pefA, iroN, cdtB, sipB, and spaN (invJ) have been shown to be required for full Salmonella virulence in a murine model (2, 3,5,7,8,11,12,16,18,20,26,27).…”

“…The PhoP-activated genes most dependent on the MgtA protein specify proteins of unknown function and, like the mgtA gene, are not required for virulence in an animal (19)(20)(21). Therefore, we hoped that the investigation of mgtA-dependent phenotypes might reveal a novel aspect(s) of Salmonella's lifestyle.…”
