Characterization of the phytochelatins and their derivatives in rice exposed to cadmium based on high-performance liquid chromatography coupled with data-dependent hybrid linear ion trap orbitrap mass spectrometry

Abstract: The HPLC/LTQ-Orbitrap method was successfully developed for the identification of PCs and their derivatives. The C-terminal linked to Gly is replaced with Glu, Ser, Asn, Gln or Cys, thereby creating a family of chemicals that share several structural properties. This technique could be particularly useful for investigators studying plant metabolomics. Copyright © 2016 John Wiley & Sons, Ltd.

“…Additional details of metabolite validation and PyC-metal complex collision-induced dissociation are provided in the Supplementary Information (Figure S-1 and Figure S-3 to S-8). Examples are provided along with literature data for PyCs and PyC-metal complexes in Table 2 (6,18,19,28–30).…”

“…The first PyC form identified was (γ-Glu-Cys) n -Gly (n = 2–11) (5). Other forms exist in which glycine (Gly) can be substituted with β-alanine (β-Ala), Ala, glutamine (Gln), serine (Ser) or glutamate (Glu) or with no additional amino acid (4,6–8). Because of their high thiol (-SH) content due to Cys residues, PyCs have strong metal-binding abilities with increasing metal capacity with increasing PyC size (3,9).…”

“…Various analytical methods are used to identify PCs and their complexes with metals [38,56,74,83,120,[152][153][154], and their advantages and disadvantages are discussed in the review by Ahmad and co-authors [155]. The most numerous studies are devoted to the role of glutathione and PCs in the detoxification of Cd, since Cd ions have the highest affinity for these ligands compared to other metals (Mn, Fe, Cu, Zn) [156].…”

“…Instead of glycine, at the C-terminus of the polypeptide there can be glutamine (Gln), glutamic acid (Glu), serine (Ser), alanine (Ala), β-alanine (β-Ala), asparagine (Asn), or cysteine (Cys), or the C-terminal amino acid may be absent. In addition, there have been found several PC derivatives that lack either Glu or Cys residues from the γ-Glu-Cys structure [45,83,86,99]. Thus, several families of PCs have been currently identified [83,110,112,165] (Table 1).…”
