Characterization of the metabolic burden on Escherichia coli DH1 cells imposed by the presence of a plasmid containing a gene therapy sequence

Abstract: The presence of a plasmid, containing gene sequences for DNA immunotherapy that are not expressed in microbial culture, imposed a degradation in bioreactor performance in cultures of the host E. coli strain. Significant decreases in growth rate (24%) and biomass yield (7%) and a corresponding increase in overflow metabolism were observed in a strain containing a therapeutic sequence (a hepatitis B antigen under the control of a CMV promotor). The observed increase in overflow metabolism was incorporated into a… Show more

“…Similar observations were reported for pWW100, inferring biphenyl metabolism (BPH ϩ ) (18), and pWWO-1, inferring toluene degradation (TOL ϩ ) (19) in P. putida. Metabolic burden related to extra DNA/plasmid replication (20,21) is suggested as a major reason for the growth retardation of plasmid-bearing cells compared to the growth of cured cells (17).…”

“…The conjugative proficiency requires the expression of set of transmembrane proteins which are known collectively as type IV secretion systems (T4SS; Lawley, Klimke, Gubbins, & Frost, 2003). The presence of conjugative plasmids and the expression of conjugative machinery are detrimental for the host cell fitness (Rozkov et al., 2004), but there is no consensus on the valid ranges of metabolic costs imposed by the conjugative process. Therefore, in this model the short‐term dynamics of two plasmid system P1 and P2 is simulated.…”

“…Besides the above mentioned regulatory aspect, Rozkov et al (2004) note another one that should be taken into consideration when selecting the plasmid stabilizing system. According to these authors, the presence of an antibiotic selection marker imposes a huge metabolic burden on an expression system.…”
