Characterization of the mechanisms involved in the gastric antisecretory effect of TLQP-21, a vgf-derived peptide, in rats

Abstract: TLQP-21, a vgf-derived peptide modulates gastric emptying and prevents ethanol-induced gastric lesions in rats. However, it remains to be studied whether or not TLQP-21 affects gastric acid secretion. In this study, we evaluated the effects of central (0.8-8 nmol/rat) or peripheral (48-240 nmol/kg, intraperitoneally) TLQP-21 administration on gastric acid secretion in pylorus-ligated rats. The mechanisms involved in such activity were also examined. Central TLQP-21 injection significantly reduced gastric acid … Show more

“…Intracerebroventricular administration of VGF-derived peptide TLQP-21 in mice increased energy expenditure and body temperature without altering circulating thyroid hormones (free T3 and T4) and prevented high-fat diet-induced obesity [41, 42]. Intracerebroventricularly injected TLQP-21 inhibited gastric emptying, decreased gastric acid secretion and reduced ethanol-induced gastric lesion in rats [43-45]. These functional activities were inhibited by cyclooxygenase (COX) inhibitors, suggesting the involvement of prostaglandins.…”

“…TLQP-21 exerts a number of biological functions including: chronic i.c.v. infusion increases energy expenditure and prevents the early phase of diet-induced obesity [12], peripheral infusion induces prolipolytic effect [13], it regulates food intake and body weight in Syrian hamsters but not in mice [14,15], it regulates gastric functions in vitro and in vivo [16,17,18], and data report the modulation of this peptide by feeding in gastric endocrine cells [19]. TLQP-21 did not show any activity on GH expression and secretion in porcine pituitary slices in vitro [20] and in vivo after chronic TLQP-21 i.v.…”

“…This peptide is known to play a role in centrally stimulating the autonomic nervous system by means of central prostaglandins (PGs), in peripherally stimulating adrenomedullary activity and in adipose tissue catabolism [2]. In addition, TLQP-21 has been shown to stimulate the in vitro rat gastric fundus contraction, and to exert an inhibitory in vivo role in the central regulation of gastric emptying and secretion through a PG-mediated mechanism [30,32]. TLQP-21 exerts a protective action on the gastric mucosa exposed to ethanol and this effect is mediated by constitutive-derived NO and PGE 2 [31].…”
