Characterization of the GlutathioneS-Transferases Involved in Styrene Degradation in Gordonia rubripertincta CWB2

Abstract: Styrene is a toxic compound that is used at a large scale by industry for plastic production. Bacterial degradation of styrene is a possibility for bioremediation and pollution prevention.

“…These data suggest that AldH is involved in the isoprene metabolic pathway subsequent to the production of HGMB. A role for AldH was previously suggested in the functionally similar styrene metabolic pathway (Lienkamp et al, 2021), in which StyH (catalyzing the equivalent reactions as IsoH) and AldH1 (equivalent to AldH in isoprene metabolism) may both catalyze the second NAD + -reducing oxidation step, equivalent to the conversion of GMB to GMBA in isoprene metabolism (Figure 3). The metabolomics data support the role of AldH in the conversion of HGMB to GMBA in Variovorax sp.…”

“…The activity of recombinant WS11-IsoI was assayed spectroscopically in a preheated Shimadzu UV-1800 spectrophotometer with a Shimadzu Constant-Temperature Measurement (CPS)-controller using 1-chloro-2,4-dinitrobenzene (CDNB) (340 nm) and 1,2-dichloro-4-nitrobenzene (DCNB) (345 nm) as adapted from Lienkamp et al ( 31 ) and van Hylckama Vlieg et al, ( 25 ). Briefly, the reactions contained 50 mM potassium phosphate buffer (pH 7.0), 10 mM glutathione, 0.5 mg mL −1 recombinant WS11-IsoI, and 1.5 mM CDNB/DCNB in a final volume of 1 mL in a quartz cuvette and were performed at 30°C.…”

“…WS11 on isoprene ( 23 ). A putative role for AldH was suggested by a study of an analogous pathway of styrene metabolism in which an AldH homologue may act alongside StyH in the conversion of (S)-1-phenyl-2-acetaldehyde glutathione to (S)-1-phenyl-2-acetic acid glutathione ( 30 , 31 ). The equivalent reaction in the isoprene metabolic pathway is the oxidation of GMB to GMBA.…”

“…Gordonia members are Gram-positive aerobic heterotrophs, of which some can degrade styrene ( 1 ). Bacteriophages that infect Gordonia may therefore be important in the exchange of bioremediation genes ( 2 , 3 ).…”
