Only bacteria sufficiently resistant to the toxic compounds in their environment can be used for the efficient biodegradation process in order to eliminate a widespread contamination by polychlorinated biphenyls (PCBs). The presence of PCBs results in bacterial controlled rigidification of cytoplasmic membrane. The four bacterial isolates from long-term PCB-contaminated soil (Alcaligenes xylosoxidans, Pseudomonas stutzeri) and sediment (Ochrobactrum anthropi, Pseudomonas veronii) have been used to select the strain most adapted to the PCBs, i.e. with efficient changes in the membrane phospholipid fatty acids. PCBs and their toxic degradation products -the 3-chlorobenzoic acids (3-CBA as the most toxic one) -were added separately to the liquid medium with glucose in two experimental sets: at lag phase and in stationary phase of bacterial growth in order to evaluate the effects of chemicals to cytoplasmic membrane. The main parameter -the changes in fatty acids composition (in the total lipids and the main membrane phospholipid phosphatidyletanolamine) were studied. 3-CBA caused growth inhibition when added at lag phase. However, when added during the stationary growth, inhibition was not observed. Similarly, after addition of PCBs to the stationary growth culture, inhibition of growth was not observed with all tested strains (except for P. stutzeri). This fact indicates the importance of time contact of bacteria during growth phase with xenobiotics. O. anthropi and A. xylosoxidans appeared to be the most adapted to the presence of PCBs (with sufficient membrane adaptation), active under the adverse conditions, and able to survive in the contaminated environment.

The effect of polychlorinated biphenyls on biomass production, lipid accumulation, and on the fatty acid profi le of the major membrane lipids of Alcaligenes xylosoxidans isolated from a soil long-term contaminated with polychlorinated biphenyls was examined. The lowest bacterial growth was observed in the presence of biphenyl and polychlorinated biphenyls. On the other hand, the highest growth stimulation was observed in the presence polychlorinated biphenyls. Higher growth ability was observed when polychlorinated biphenyls or 3-chlorobenzoic acid were added after three days of cultivation. Higher lipid accumulation corresponded with lower growth ability. The importance of time when bacterial strain meets polychlorinated biphenyls or 3-chlorobenzoic acid should be emphasized. The addition of toxic organic compound later in stationary growth phase led to better survival and evoked lower membrane adaptation. However, polychlorinated biphenyls or 3-chlorobenzoic acid amendment at the beginning of cultivation stimulated adaptation responses as membrane saturation, cis to trans isomerisation, and branched fatty acids alteration and decreased growth ability. Palmitic acid and 17-cyclopropyl fatty acid were observed as prevalent in the bacterial membrane as well as in the total lipids.

Bioaccumulation of six PCB indicator congeners in a heavily polluted water reservoir in Eastern Slovakia: tissue-specific distribution in fish and their parasites

Concentrations of six indicator PCB congeners (IUPAC nos. 28, 52, 101, 138, 153, and 180) were measured in several organs and adipose tissue of a freshwater predatory fishes (European perch, northern pike, pike perch, wels catfish) as well as in nonpredators (common carp, freshwater bream, goldfish, white bream) and in acanthocephalan Acanthocephalus lucii from the water reservoir Zemplínska šírava (Eastern Slovakia), which is considered to be one of the most PCB-contaminated places in Europe. Concentration of PCBs was determined by capillary gas chromatography in samples from May to September 2009. The two-way main-effect ANOVA confirmed that feeding habits of fish (P < 0.00001) and peculiarity of individual fish organs (P < 0.01) affect PCB bioaccumulation. The total amount of PCBs was significantly higher (P < 0.05) in predators compared to nonpredators. Tissue-specific differences were found in PCB accumulation in both fish groups. PCBs were predominantly accumulated in the liver and hard roe. Individual congeners were not distributed homogeneously within the investigated organs and adipose tissue. PCB 153 was present in higher concentrations than the other congeners in all fish organs as well as in adipose tissue comprising an average 31 and 34 % of ΣPCB in predators and nonpredators, respectively. Acanthocephalans, attached to the intestine of perch, absorbed significantly higher concentrations of PCBs (P < 0.001) than the muscles, liver, kidney, brain, and adipose tissue of their host. About 20 times lower amount of PCBs was detected in the liver and almost 3 times in muscles of infected perch. Data on PCB accumulation in perch infected with acanthocephalans demonstrated a decline of PCB values in all organs as well as in adipose tissue compared to noninfected fish. About 20 times lower amount of PCBs was detected in the liver and almost 3 times in muscles of infected perch. Present results could indicate that some parasitic organisms may influence positively their hosts in PCB-contaminated environment.
