Characterization of the bla KPC-2 and bla KPC-3 genes and the novel bla KPC-15 gene in Klebsiella pneumoniae

Abstract: Three Klebsiella pneumoniae isolates exhibiting high-level resistance to carbapenem were analysed by PCR, PFGE, gene mapping, plasmid conjugation and Southern blot hybridization using a bla KPC probe. In addition to the frequently reported bla KPC-2 and bla KPC-3 genes, a novel bla KPC-15 gene was identified in one of the isolates. The results of plasmid analysis and Southern blot hybridization revealed that the three bla KPC genes were located on transferable plasmids exhibiting three different patterns. The … Show more

“…The transformant was also much less resistant than the original isolate, it was most likely due to the lack of any permeability lesion. The genetic environment of the bla KPC-15 gene in the plasmid showed a structural and context similarity to those characteristics found in plasmid pKP048 [33], except that a truncated bla TEM-12 gene and the bla KPC-15 variant were inserted and located between IS Kpn8 and the novel bla KPC-15 variant [24]. The nucleotides upstream of the bla KPC-15 translational start codon were also shown in Figure 3.…”

“…There were 5 different genes in the sequence, including the genes tnpR , bla TEM-12 and bla KPC-15 , and the transposons IS Kpn8 and IS Kpn6 -like [24]. The sites of the transcriptional promoters for the bla TEM-12 and bla KPC-15 genes were indicated as +1 and the −10 and −35 regions were also shown; however, there were no obvious transcriptional promoters in tnpR , IS Kpn8 and IS Kpn6 -like transposons.…”

“…These enzymes hydrolyze a wide spectrum of β-lactams, including penicillins, cephalosporins, and carbapenems [3]. The bla KPC gene possesses different variants (KPC-2 to KPC-15) that are sporadically found in other species, such as non-fermenting bacteria [47]. …”
