Characterization of Temperate Actinophage φC31 Isolated from Streptomyces coelicolor A3(2)

Abstract: Actinophage 4C31 isolated from Streptomyces coelicolor A3(2), the only strain among actinomycetes for which a genetic map had been constructed, appears to be a typical temperate phage. After 4C31 infection, true lysogenic cultures arose which liberated phage and were immune to infection with homologous phage after repeated single-colony isolations and treatment with phage-specific antiserum. Clear-plaque (c) mutants were derived from q5C31 phage which failed to lysogenize sensitive cultures. Actinophage 45C31 … Show more

“…Materials and methods S. lividans 66 (Lomovskaya et al, 1972) (John Innes stock number 1326) was the host for 4C31 propagation and assay (Chater et al, 1981) and for transfection of protoplasts (Rodicio and Chater, 1982). S. coelicolor J1507 , containing plasmid SCP1 integrated stably into the chromosome in the NF mode , was the usual host for mutational cloning.…”

“…Bacterial strains, plasmids and growth conditions Streptomyces lividans prototrophic strain 1326 (Lomovskaya et al, 1972) was grown on NE agar plates (Murakami et al, 1986) or YEME liquid medium containing 34% sucrose (Hopwood et al, 1985). Resistance to thiostrepton was provided by tsr cloned in the streptomycete cloning vector p1I61 (Thompson et al, 1982b).…”
