At least 17 kb of DNA from the large unisolatable Streptomyces coelicolor A3(2) plasmid SCP1 are concerned with methylenomycin biosynthesis. Mutational cloning analysis, using insert‐directed integration of att site deleted phage vectors into an SCP1‐containing host, provided evidence of two large transcription units, of at least 6.6 kb and 9.5 kb. At the leftmost apparent end of the larger (left‐hand) transcription unit is a region apparently involved in negative regulation of methylenomycin biosynthesis: when fragments from this region were used to direct phage integration, marked overproduction of methylenomycin resulted. The methylenomycin resistance determinant is located at the rightmost end of this same transcription unit. Hybridisation analysis with 13 kb of the cloned mmy region showed that it was closely similar to a segment of pSV1, a plasmid that specifies methylenomycin biosynthesis in S. violaceus‐ruber SANK 95570.

Although the antibiotic thiostrepton is best known as an inhibitor of protein synthesis, it also, at extremely low concentrations (< 10-9 M), induces the expression of a regulon of unknown function in certain Streptomyces species. Here, we report the purification of a Streptomyces lividans thiostrepton-induced transcriptional activator protein, TipAL, whose N-terminus is similar to a family of eubacterial regulatory proteins represented by MerR.TipAL was first purified from induced cultures of S.lividans as a factor which bound to and activated transcription from its own promoter. The tipAL gene was overexpressed in Escherichia coli and TipAL protein purifi'ed in a single step using a thiostrepton affinity column. Thiostrepton enhanced binding of TipAL to the promoter and catalysed specific transcription in vitro.TipAs, a second gene product of the same open reading frame consisting of the C-terminal domain of TipAL, is apparently translated using its own in-frame initiation site. Since it is produced in large molar excess relative to TipAL after induction and also binds thiostrepton, it may competitively modulate transcriptional activation.

Summary High‐resolution SI nuclease mapping of mRNA synthesised in vivo, in vitro run‐off transcription with RNA polymerase from Streptomyces lividans and gene fusions were used to analyse the transcriptional organization of the Sall restriction‐modification system of Streptomyces albus G. The sallR and sallM genes that encode the restriction endonuclease and its cognate methyltransferase constitute an operon which is mainly transcribed from sal‐pR1, a promoter located immediately upstream of saIIR, with two possible minor promoters further upstream. Another promoter, sal‐pM, is within the 3′ end of the saIIR coding region, and allows expression of the modification gene in the absence of sal‐pR1. The sal‐pM promoter might be involved in the establishment of modification prior to restriction endonuclease activity. Sequences upstream of the apparent transcriptional start sites for sat‐pR1 and sal‐pM show similarity with the −10 region of typical vegetatively expressed eubacterial promoters, but appropriately centered −35 regions are absent.
