Characterization of Some Commercially Important Octopus (Mollusca: Cephalopoda) from Indonesian Waters using Mitochondrial DNA Cytochrome Oxidase Sub-Unit I (Mt-DNA COI)

Abstract: As per the FAO data, octopus identification is very limited in the species level at world fishery and also they are cryptic nature. On the other hand, Indonesia is one of the top ten highest octopus exporters. This study therefore aimed to determine the species of octopus based on phylogenetic analysis of mt-DNA COI. Octopuses were collected from nine different locations throughout Indonesia, i.e., Anambas, Bangka-Belitung, Cirebon, Karimunjawa, Tuban, Lombok, Buton, Wakatobi and Jayapura. Samples were mostly … Show more

“…DNA extraction using Chelex 10%, which aims to obtain sample DNA by isolating mtDNA in the sample (Walsh et al, 1991). The sample was heated at a state the exact temperature, in the presence of the resin contained in Chelex products that can be used to extract DNA (Marwayana, 2015;Kholilah et al, 2021;Putri et al, 2022). Sample tissue was taken ±2 mm and then inserted into a tube containing 10% Chelex solution.…”

“…This has led to molecular identification becoming an alternative, particularly in Indonesia. Molecular identification is easier to learn and can be performed by anyone for research on cryptic organisms like fish (Nursalim et al, 2022), Decapoda (Cheng et al, 2020;Ambariyanto et al, 2023), Coral (Insafitri et al, 2023), Rajungan (Marita et al, 2023) and Cephalopoda (Kholilah et al, 2021a).…”

“…Coral reef is a coastal ecosystem which contains important and beneficial natural resources. Coral reefs have many good functions in ecology that act as biota habitats for many biotas such as octopus (Kholilah et al 2021), giant clam (DeBoer et al 2012), sea cucumber (Widianingsih et al 2021;Sulardiono et al 2022), and lobsters (Haryono and Ambariyanto 2018). Even the research of Pertiwi et al (2015), states that dead corals can become habitats for decapods.…”
