Characterization of somatostatin in peripheral and portal plasma in the duck: in-vivo metabolism of somatostatin-28 and-14

Abstract: A sensitive and specific radioimmunoassay without an extraction step was developed for somatostatin in duck plasma. Degradation of Tyr1-125I-labelled somatostatin-14 (S-14) averaged 2% for blood collected with EDTA and zymofren. Recovery of somatostatin-like immunoreactivity (SLI), added to the plasma, averaged 91% for S-14 and 86% for S-28. Chromatographic analysis of portal plasma on Sephadex G-25 showed three peaks: one peak coeluted with cytochrome c (mol. wt 12500) in the void volume and was called 'big' … Show more

“…Pancreatic somatostatin was assayed by the RIA de¬ scribed by Discala-Guénot et al (1984) using a rabbit anti-somatostatin 14 serum and synthetic somatostatin 14 as standard. The sensitivity of the assay was 10 ng/1 and inter-and intra-assay variation averaged 8.9 and 4.9%, respectively.…”

“…wt 1600; Beckman). Hormone analysis Somatostatin was estimated in pancreatic extracts and column fractions by radioimmunoassay (Di Scala-Guenot et al 1984), using somatostatin-14 and somatostatin-28 as standards and '^I-TyrMabelled somatostatin-14 (New England Nuclear, Dreieich, F.R.G.) as the tracer.…”

“…Somatostatin-like immuno¬ reactivity (SLI) has similarly been located in birds, particularly in the pancreas , intestine (SLI-14 and SLI-28) and plasma ('big' SLI) of adult ducks (Di Scala-Guenot, Strosser & Mialhe, 1984.…”

“…In ducks, somatostatin is present in the pancreas, intes¬ tine and plasma (Di Scala-Guenot, Strosser & Mialhe, 1984 and in the hypothalamus . In vivo, somatostatin-14 (S-14) stimu¬ lates pancreatic glucagon release and inhibits insulin secretion in adult and young ducks (Strosser, Cohen, Harvey & Mialhe, 1980), and in young but not in adult ducks, both S-14 and S-28 inhibit basal growth hormone (GH) secretion (Strosser, Harvey, Foltzer & Miahe, 1984.…”
