Foodborne diseases represent a major risk to public health worldwide. In this study, LPST153, a novel Salmonella lytic phage with halo (indicative of potential depolymerase activity) was isolated by employing Salmonella enterica serovar Typhimurium ATCC 13311 as the host and had excellent lytic potential against Salmonella. LPST153 is effectively able to lyse most prevalent tested serotypes of Salmonella, including S. Typhimurium, S. Enteritidis, S. Pullorum and S. Gallinarum. Morphological analysis revealed that phage LPST153 belongs to Podoviridae family and Caudovirales order and could completely prevent host bacterial growth within 9 h at multiplicity of infection (MOI) of 0.1, 1, 10 and 100. LPST153 had a latent period of 10 min and a burst size of 113 ± 8 PFU/cell. Characterization of the phage LPST153 revealed that it would be active and stable in some harsh environments or in different conditions of food processing and storage. After genome sequencing and phylogenetic analysis, it is confirmed that LPST153 is a new member of the Teseptimavirus genus of Autographivirinae subfamily. Further application experiments showed that this phage has potential in controlling Salmonella in milk and sausage. LPST153 was also able to inhibit the formation of biofilms and it had the ability to reduce and kill bacteria from inside, including existing biofilms. Therefore, the phage LPST153 could be used as a potential antibacterial agent for Salmonella control in the food industry.

Salmonella species are causing high morbidity and mortality rates in broiler chickens and other birds particularly young ages resulting in great economic losses. The present study was performed to detect the effect of age and season on isolation rates of Salmonella species from 120 examined broiler chickens with different ages (1-7 days-old and more than 7days-old) in different seasons (summer, winter, autumn and spring). The highest isolation rates of Salmonella species were obtained from broilers of 1-7 daysold in summer (40%) followed by broilers of more than 7 days old in summer (33.3%). The increased isolation rates of Salmonella species was detected from liver of broilers of both ages (1-7 and more than 7 days old). Seventeen Salmonella isolates were obtained in this study from both ages. Serotyping of these Salmonella isolates revealed S. Enteritidis (5 isolates), S. Typhimurium (3 isolates), S. Infantis, S. Bargny, S. Newport, S. Magherafelt (2 isolates) and finally S. Apeyeme in (1 isolate). The most frequent resistance phenotypes were tetracycline (70%-71.4%), from 1-7 days-old and more than 7 days-old chicks and finally ampicillin (60%, 57.1%). From the result obtained by using VITEK2 system, the highest resistance was recorded against Tetracycline in all Salmonella species. All Salmonella isolates showed-associated genes. The invA gene was identified in all Salmonella serovars isolated from chickens of the two age groups, whereas, avrA gene was detected in all the isolates except S. Magherafelt recovered from the age group of more than 7 days-old birds. The tetracycline resistance gene (tetA) was identified in all the examined isolates.

BackgroundSalmonella is a very important food-borne pathogen causing illness in humans. The emergence of drug-resistant strains also constitutes a serious worry to global health and livestock productivity. This study investigated Salmonella isolates from poultry and poultry products using the phenotypic antimicrobial screening as well as the molecular characteristics of Salmonella isolates. Upon serotyping of the isolates, the antimicrobial susceptibility profiling using a panel of 9 commonly used antimicrobials was done. Subsequently, the molecular profiles of all the isolates were further determined using Pulsed Field Gel Electrophoresis (PFGE) and the Whole Genome Multi-Locus Sequence Type (wgMLST) analysis in order to obtain the sequence types. ResultsThe PFGE data was input into FPQuest software, and the dendrogram generated was studied for possible genetic relatedness among the isolates. All the isolates were found to belong to the S. Enteritidis serotype with notable resistance to tetracycline, gentamycin, streptomycin, and sulfadimidine. The S. Enteritidis isolates tested predominantly subtyped into the ST11 and ST1925, which was found to be a single cell variant of ST11. The STs were found to occur in chicken meat, food, and live chicken cloacal swab, which may indicate the persistence of the bacteria in multiple foci. ConclusionThe data demonstrate the presence of S. Enteritidis among chicken, indicating its preference and reservoir status for enteric salmonella pathogens.
