Loquat is an important commercial specialty fruit tree crop in China, and as a result, there is significant interest in its genetic improvement. Wild loquat, native to China, provides a remarkable genetic resource for breeding improved varieties. Guizhou Province in southwest China contains many natural populations of loquat in its rural areas. In the present study, we analyzed the genetic diversity of native loquat growing throughout Guizhou Province together with wild relatives and domesticated loquat using a set of 20 informative SSR markers. Molecular analysis identified four gene pools, with gene pool I representing cultivated genotypes and gene pool IV containing wild relatives and native surveyed accessions. Approximately 10.8 % of the samples collected from Guizhou Province are feral forms derived from cultivated genotypes, whereas 24.9 % are most closely related to wild species. The remaining 64.3 % of the gene pool is intermediate genotypes derived from spontaneous crosses between wild native forms and cultivated loquat.

Portugal has a long tradition in viticulture and a great number of grapevine cultivars. To analyze the genetic relations among wild vines from Portuguese populations and old Portuguese grapevine cultivars we use morphological traits and chloroplastidial microsatellites from 53 accessions of four distinct populations of Vitis vinifera L. ssp. sylvestris (Gmelin) and 57 accessions of Vitis vinifera L. ssp. vinifera from the Portuguese National Ampelographic Collection. Principal coordinate analyses with the scores obtain from the descriptors of both the accessions of sylvestris and vinifera vines revealed two groups. One group is formed by the wild vine population of Alcácer do Sal and three vinifera accessions Rufete, Seara Nova and Trincadeira das Pratas and a second group includes all the other wild vines and grapevine cultivars. A total of four different chlorotypes (A, B, C and D) are present in the vinifera accessions and two in the sylvestris accessions (A, B). Chlorotype A is the most frequent in all the plants analyzed and correspond to 75.4% of the grapevine cultivars and 66% of the wild vines. The mixed distribution of chlorotypes in the Portuguese cultivars and the predominance of chlorotype A both in its wild populations and cultivars reinforced the hypothesis that West Europe was a domestication center for Vitis vinifera L. ssp. vinifera.

A survey of Vitis vinifera ssp. sylvestris (Gmeli) Hegi in Portugal has revealed the existence of wildvine populations that occur only in riparian wood habitats on river margins, as is the case for other European populations. The genetic diversity of four populations has been evaluated using nuclear and chloropastidial microsatellites as molecular markers. An analysis of molecular variance (AMOVA), showed that most of the genetic diversity was attributable to differences among individuals within populations. Only 7% of the total variance was attributable among populations; suggesting the existence of a low level of population differentiation. Chloroplastidial microsatellites revealed the expected situation for the Iberian Peninsula, (i.e. the presence of only chlorotypes A and B; with chlorotype A as the most frequent within the wild-vine populations). The diversity obtained is a starting point for the management and conservation of wild-vines in situ and ex situ. Several measurements have to be taken to maintain their natural habitat, and in order to preserve its diversity.
