Characterization of phosphate efflux pathways in rat liver mitochondria

Abstract: ATP hydrolysis catalysed by the H+-ATPase of intact mitochondria can be induced by addition of ATP in the presence of valinomycin and KCl. This leads to an increase in intramitochondrial Pi and therefore allows investigation of potential Pi efflux pathways in intact mitochondria. Combining this approach with the direct measurement of both internal and external Pi, we have attempted to determine whether Pi efflux occurs via an atractyloside-sensitive transporter, by the classical operation of the Pi/H+ and Pi/d… Show more

“…Functionally linked to ANT are the P i C [16] and, in muscle tissues, the MtCK (reviewed in [17]). To allow stoichiometry and electroneutrality of the exchange, co-transport of P i and H + is required.…”

“…The major transport system for the translocation of P i across the mitochondrial inner membrane is the mersalyl-sensitive P i C [16]. T $ -dependent P i C expression was measured in heart, kidney, liver and brain by Northern blotting with a fragment of the bovine P i C cDNA.…”

“…The protein content of mitochondria suspension was quantified by the biuret method with addition of 1% collate. 24 The mitochondrial respiratory function was measured immediately after isolation by polarographic analysis at 30ºC. Briefly, mitochondria (1 mg) were added to 1.4 ml of solution containing 250mM sucrose, 8.5mM KH 2 PO 4 , 10mM EDTA, 2mg/ml BSA and 10mM Tris-HCl, pH 7.4.…”

“…Ischemia/reperfusion injury also activates complement, cytokines, and chemokines, which are cytotoxic themselves, but also attract leukocytes into the ischemic area to cause further damage. [20][21][22][23][24] The aim of this research was to evaluate the influence of ischemia/reperfusion injury on renal compensatory growth (CGR) and mitochondrial function.…”
