Aim: Morpho-molecular analyses for taxonomic characterization of nine predominant Penicillium species present in the soil of different parts of India. Methodology: Fifteen Penicillium isolates were isolated from the soil samples collected from the experimental field of ICAR-Indian Agricultural Research Institute (IARI), New Delhi. Another twenty-six isolates were procured from Indian Type Culture Collection (ITCC), Division of Plant Pathology, ICAR-IARI, New Delhi which were isolated from the soil of different parts of India. Total 41 isolates were characterized following distinct macroscopic (colony texture, colony colour exudate production; soluble pigmentation; reverse coloration and mycelial growth) and microscopic observations (type of penicillus; shape of phialides; conidial shape, size and pigmentation). Molecular characterization was done using partial β-tubulin gene sequence which is considered an excellent marker in differentiating Penicillium species. Results: The morphological data in species description perfectly matched with molecular data generated using β-tubulin gene marker and nine different species viz., P. aethiopicum, P. chrysogenum, P. crustosum, P. janthinellum, P. mononematosum, P. oxalicum, P. polonicum, P. singorense and Talaromyces pinophilus (Syn. P. pinophilum) were confirmed. Interpretation: Above nine Penicillium species are found predominantly in the soil collected from different parts of India. The β-tubulin gene can be considered as an excellent marker to differentiate Penicillium species as confirmed from this study. The combined morpho- molecular analyses can be further utilized to delineate morphologically similar Penicillium species and also helpful to establish new species of Penicillium.

Table grapes are exposed to fungal infections before and after harvest. In particular, Aspergillus, Penicillium, and Alternaria can cause decays and contamination by mycotoxins. The main fungi affecting Lebanese table grapes after harvest were assessed as epiphytic populations, latent infections, and rots. Effects of storage with and without SO2 generating pads were also evaluated. Representative isolates of toxigenic genera were characterised, and their genetic potential to produce ochratoxin A, fumonisins, and patulin was established. The epiphytic populations mainly included wound pathogens (Aspergillus spp. and Penicillium spp.), while latent infections and rots were mostly caused by Botrytis spp. The use of SO2 generating pads reduced the epiphytic populations and rots, but was less effective against latent infections. Characterization of Aspergillus, Penicillium, and Alternaria isolates showed that A. tubingensis, P. glabrum, and A. alternata were the most common species. Strains of A. welwitschiae and P. expansum were also found to be genetically able to produce, respectively, ochratoxin A plus fumonisins and patulin. These data demonstrate the need for effective measures to prevent postharvest losses caused by toxigenic fungi.

Pomegranate (Punica granatum L.) is an emerging crop in Italy and particularly in southern regions, such as Apulia, Basilicata, and Sicily, due to favorable climatic conditions. The crop is affected by several pathogenic fungi, primarily in the field, but also during postharvest phases. The most important postharvest fungal diseases in pomegranate are gray and blue molds, black heart and black spot, anthracnose, dry rot, and various soft rots. The limited number of fungicides allowed for treatment in the field and the lack of postharvest fungicides make it difficult to control latent, quiescent, and incipient fungal infections. Symptomatic pomegranates from southern Italy were sampled and isolated fungi were morphologically and molecularly characterized. The data obtained revealed that various species of Penicillium sensu lato (including Talaromyces genus), Alternaria spp., Coniella granati, and Botrytis cinerea were the principal etiological agents of postharvest pomegranate fruit diseases; other relevant pathogens, although less represented, were ascribable to Aspergillus sect. nigri, Colletotrichum acutatum sensu stricto, and Cytospora punicae. About two thirds of the isolated pathogens were responsible for latent infections. The results obtained may be useful in planning phytosanitary control strategies from the field to storage, so as to reduce yield losses.
