IgG, IgM and IgA antibody responses against Micropolyspora faeni (Mf) antigens were studied by means of immunoblotting experiments using 70 sera derived from three groups of farmers, namely patients with extrinsic allergic alveolitis (EAA) due to thermophilic actinomycetes (n = 25), patients without EAA but with hay exposure (n = 14), and patients suspected to have EAA (n = 31), and 27 sera from two groups of control persons (healthy laboratory workers, n = 13; healthy farmers, n = 14). Patients with EAA showed IgG, IgM and IgA antibody responses mainly against the antigens with molecular weights (MW) of 11, 12, 25, 35 and 60 kD ("major antigens"), and in addition, but less often, against six antigens with MW in the range of 15 to 62.5 kD ("minor antigens"). The other two groups of patients and also the exposed control persons showed very similar results; however, the antibody response in healthy farmers was substantially weaker in comparison to the three groups of patients and was almost limited to the major antigens with MW 11, 25 and 60 kD. Although patients with proven EAA had higher amounts of antibodies, there was no correlation between this antibody response and the onset of disease. The results indicate the necessity of including at least the major antigens with MW of 11, 25 and 60 kD in all extracts used for in vitro diagnosis of Mf-induced EAA.

Zusammenfassung: Die Farmerlunge ist eine Hypersensivitäts‐Pneumonitis, die in einer akuten und chronischen Form meistens bei Landwirten in regenreichen Gebieten vorkommt. Sie hängt mit der Verfütterung von ungenügend getrockneten, schimmligen Futtermitteln zusammen, bei der pflanzlicher Staub eingeatmet wird, der große Mengen von Sporen thermophiler Aktinomyzeten enthält. Die Krankheitserscheinungen beruhen auf einer toxischen Pneumonie durch Immunreaktionen im periphären Teil des bronchopulmonalen Systems, die bei Kontakt mit inhalierten Allergenen entstehen. Dasselbe klinische und röntgenologische Bild wurde in der ČSSR bei einigen Gruppen von jungen Leuten beobachtet, die einmalig in der Landwirtschaft ausgeholfen hatten, weiter bei Leuten, die bei der industriellen Verabreichung von A. niger tätig waren, sowie auch bei Landwirten, die mit der Fütterung und Futterzubereitung beschäftigt waren. Die Diagnose konnte auf Grund der serologischen und allergologischen Untersuchung sowie auch der Kultivationsergebnisse getroffen werden. Diese Erkrankung, die wir als ein der Farmerlunge ähnliches Syndrom benannten, wurde von Penicillien und Aspergillen verursacht. Die pathogenetische Wirkung dieser Luftpilze auf den Respirationstrakt wurde durch Experimente an Versuchstieren bestätigt. Summary: Farmer's lung is a form of hypersensitivity pneumonitis that occurs in an acute and in a chronic form, especially in a farmers in rainy countries. It is connected with feeding of damp mouldy hay or fodder when the workers inhale plant dust containing a great quantity of spores of thermophilic actinomycetes. The symptoms are based on a toxic pneumonitis caused by immunoreactions in the periphery of the bronchopulmonal systems that occur after contact with the inhaled allergens. The same clinical and radiological picture was observed in Czechoslovakia in some groups of young people working only one time as farmer's help, then in industrial workers cultivating A, niger and in farmers feeding and preparing the fodder. The diagnosis was made on the basis of serological and allergological investigations as well as on results of cultivation. This disease that we named as a Syndrom Resembling Farmer's Lung was caused by Penicillia and Aspergilli. The pathogenic effect of these air‐borne fungi on the respiratory tract was confirmed by experiments in rabbits.

A novel method for permanent imprints from dye‐stained electrophoretograms

A simple method for obtaining permanent imprints of Coomassie Blue and Amido Black stained electrophoretograms on developed unexposed photographic paper and film is described. Copies of slab and two-dimensional electrophoretograms and crossed immunoelectrophoretograms were obtained by this technique. The method is fast, simple, inexpensive and can be adapted to make as many as five copies by repeated staining of the electrophoretogram. The copies are the exact size of the original gels and can be stored for a long time. The possible use of these imprints for the comparative study of immunoblots with the corresponding protein-stained gels is discussed.
