Characterization of lymphoblast mitochondria from patients with Barth syndrome

Abstract: Barth syndrome (BTHS) is a multisystem disorder of individuals who carry mutations in tafazzin, a putative phospholipid acyltransferase. We investigated the hypothesis that BTHS is caused by specific impairment of the mitochondrial lipid metabolism. The fatty acid composition of all major mitochondrial phospholipids, phosphatidylcholine (PC), phosphatidylethanolamine (PE), and cardiolipin (CL), changed in lymphoblasts from BTHS patients. These changes were most extensive in CL and least extensive in PE. The co… Show more

“…Similar results were obtained after 24 h of 32 Pi incorporation (data not shown). Alteration in the fatty acid composition of CL was observed in BTHS cells (Table 3), as reported previously (44,46). Despite the low number of subjects to perform accurate statistical analysis, we observed that C18:2 n-6 and C20:3 n-6 were decreased dramatically with a concomitant increase of the saturated fatty acids C16:0 and C18:0.…”

“…Epstein-Barr virus-transformed lymphoblasts from control subjects and patients with BHTS (44) were cultured in suspension in RPMI 1640 medium in the presence of 10% (v/v) fetal bovine serum, 100 U/ml penicillin, and 100 mg/ml streptomycin at 37jC in a 5% CO 2 atmosphere as described previously (46).…”

“…Although tetralinoleoyl-cardiolipin was the sole species in complex IV and the dominant species in the ADP-ATP carrier, an absolute requirement for symmetric cardiolipin cannot be postulated, because these proteins remain active in tafazzin mutants, in which symmetric cardiolipin virtually disappears (123,124).…”

“…Up to now, CL and MLCL analyses in BTHS, with or without diagnostic intent, have been set up on lymphoblasts ( 18 ), lymphocytes ( 19 ), platelets (20)(21)(22), fi broblasts ( 23,24 ), neutrophils ( 25 ), leukocytes ( 26 ), bloodspots ( 27 ), and tissues ( 19 ). All these reports, including the validated methods ( 26,27 ), are based on lipid extraction followed by HPLC and ESI/MS analysis.…”
