Characterization ofXenopus laeviscDNA clones of the c-ets-1proto-oncogene

Abstract: FFSCYT ME GQEYP APL Two XI-ets-J gene transcripts of 4.4 kb (major species) and 7.5 kb (minor species) were detected in the poly(A)+ fraction from the early stages of the oogenesis to the late stages of embryogenesis. They appear to decline regularly during embryogenesis. They were not at all detected in several adult organs, except in spleen where the larger transcript (7.5 kb) is the major one.

“…Several ETS family genes, including Ets1, Fli and Erg, are known to be expressed in Xenopus embryonic endothelial cells (Stiegler et al, 1990;Stiegler et al, 1993;Meyer et al, 1993;Meyer et al, 1995;Meyer et al, 1997;Baltzinger et al, 1999). For example, whole-mount in situ hybridization for Flk1 and Erg transcripts (Fig.…”

“…In order to get a deeper insight into the function of this family of genes during early development and since mis-and overexpression of a gene can easily be followed in Xenopus laevis embryos, we have previously isolated and characterized the Xenopus laevis cDNAs of the Xl ets-1, Xl ets-2, and Xl fli genes (Stiegler et al, 1990;Wolff et al, 1991;Meyer et al, 1993;Stiegler et al, 1993;Meyer et al, 1995Meyer et al, , 1997. While Xl ets-1 and Xl ets-2 are maternally expressed, Xl fli is expressed only zygotically, from neurulation to late stages of embryogenesis.…”

“…mRNAs coding for two highly related ets family members, Ets-1 and Ets-2, were identified inX. laevis oocytes (9,11,40,49 Neither of these antibodies had a specific effect on the mobility of the C1 and C2 complexes formed by XrpFI (data not shown).…”

“…Potential Ets binding sites are found in the promoters of genes involved in the early response to growth stimuli (16,46) and genes that are highly expressed in transformed cells (47), but most of the target genes for the Ets regulators are still unknown. The tissue distribution of known ets domain-expressed proteins suggests that they could regulate the expression of tissuespecific genes (21,36,40,41,50 (12). Embryos, a generous gift from Paola Pierandrei-Amaldi of Consiglio Nazionale delle Richerche Cellular Biology Laboratory of Rome, were obtained as described by Brown and Littna (7) and were grown at 22°C in dechlorinated water and staged as described by Nieuwkoop and Faber (31).…”
