Clay–polymer nanocomposites have exhibited a great potential as carriers for controlled release drug delivery. This study aims to prepare exfoliated montmorillonite–Eudragit RS nanocomposites using reactive melt extrusion and investigate the influence of claying loading, clay types (sodium montmorillonite (Cloisite Na) vs. organomodified montmorillonite (Cloisite 20)) on clay–polymer interactions and drug release properties. The clays were used as the filler material at various levels in Eudragit RS and theophylline was used as the active pharmaceutical ingredient. The resulting structure of the nanocomposites was characterized using TEM (transmission electron microscopy) and XRPD (X-ray powder diffraction). The hygroscopicity of the nanocomposites was investigated using DVS (dynamic vapor sorption). The effect of the interfacial interaction between the polymer and clay sheet, the clay loading as well as the clay type on the drug release behavior were further studied by dissolution testing. TEM and XRPD data show that when the clay content is increased from 5% to 15% by weight, the nanocomposite’s structure switches from a fully exfoliated state to intercalated structures or partial exfoliation with stacked clay layers. FT-IR (fourier transform infrared spectroscopy) and ssNMR (solid-state NMR) results suggest that Cloisite Na and Cloisite 20 layers exhibit different interaction strengths with polymer networks by creating compacted complex structures. The addition of nanoclay in the formulation could robustly adjust drug release profiles, and the clay concentration and type are important factors that affect the crossing-linking density of the nanocomposites by adjusting the drug release properties. This study indicates that the clay–Eudragit RS nanocomposites provide an improved oral controlled drug delivery system that minimizes the drug dosing frequency, potentially leading to improved patient compliance.

The study of pharmacological interactions between herbal remedies and conventional drugs is important because consuming traditional herbal remedies as supplements or alternative medicine is fairly common and their concomitant administration with prescribed drugs could either have a favorable or unfavorable effect. Therefore, this work aims to determine the pharmacological interactions of a turmeric acetone extract (TAE) and its main metabolite (curcumin) with common anti-ulcer drugs (ranitidine and bismuth subsalicylate), using an ethanol-induced ulcer model in Wistar rats. The analysis of the interactions was carried out via the Combination Index-Isobologram Equation method. The combination index (CI) calculated at 0.5 of the affected fraction (fa) indicated that the TAE or curcumin in combination with ranitidine had a subadditive interaction. The results suggest that this antagonistic mechanism is associated to the mucoadhesion of curcumin and the TAE, determined by rheological measurements. Contrastingly, both the TAE and curcumin combined with bismuth subsalicylate had an additive relationship, which means that there is no pharmacological interaction. This agrees with the normalized isobolograms obtained for each combination. The results of this study suggest that mucoadhesion of curcumin and the TAE could interfere in the effectiveness of ranitidine, and even other drugs.

In this work, curcumin was spray-dried (SD) with Aloe vera mucilage as the encapsulating agent giving particular attention to the effect of the relevant process parameters (feed flow rate, atomization speed, and inlet air temperature) on the resulting properties of the SD powders.The powders obtained were analyzed by infrared spectroscopy, scanning electron microscopy, rheology, and release profiles. Results show that the best drying conditions are low SD inlet air temperature (150 C), low feed flow rate (1.5 L/hr), and high atomization speed (27,500 rpm).These conditions produced particles with smooth morphologies, preserving the total phenolic content (0.0611 μg EAG), with a radical scavenging capacity of 911.48 μmol Trolox/mg. Release profiles revealed an extended release of encapsulated material with a maximum of about 65% at 24 hr for these conditions and point out the suitability of encapsulated systems for potential applications in functional foods, antioxidant systems, pharmaceutical, and organic pigments. Practical applicationsThis research presents an optimization method to obtain powders of mucilage (Aloe vera, Av)curcumin (Cu) by spray drying (SD), where the Av was used as wall material to encapsulate Cu and obtain the best process conditions (SD) to produce prolonged delivery systems of antioxidant compounds that can be used in the food and pharmaceutical industry. The encapsulated systems were analyzed by rheological tests and were supported with other characterizations, such as antioxidant capacity, morphology (SEM), chemical (FT-IR), and releasing profiles.
