Characterization of Human Cellular Immune Responses to NovelMycobacterium tuberculosisAntigens Encoded by Genomic Regions Absent inMycobacterium bovisBCG

Abstract: Comparative genomics has identified several regions of differences (RDs) between the infectious Mycobacterium tuberculosis and the vaccine strains of Mycobacterium bovis BCG. We aimed to evaluate the cellular immune responses induced by antigens encoded by genes predicted in 11 RDs. Synthetic peptides covering the sequences of RD1, RD4 to RD7, RD9 to RD13, and RD15 were tested for antigen-induced proliferation and secretion of Th1 cytokine, gamma interferon (IFN-␥), by peripheral blood mononuclear cells (PBMC)… Show more

“…But among all mce3 operon proteins, only region of difference 1504 (Mce3A) was demonstrated to be able to induce a strong Th1 bias in healthy subjects and a weak Th1 bias in TB patients, whereas other proteins of mce3 operon induced only moderate to weak Th1 biases in both TB patients and healthy subjects (24), suggesting that other proteins among mce3 operon might have potential immune-modulating effects. Despite the potential function of Mce proteins on intracellular survival of M. tuberculosis, little progress has been made to define the specific functional mechanisms of each Mce protein.…”

“…The results have suggested the presence of immunodominant antigens in RD7 and RD9 [49][50][51][52][53]. However, peptide pools of these RDs were also recognized in the same assays by cells from M. bovis BCG-vaccinated healthy subjects [49,53]. Further testing of the individual proteins present in RD7 and RD9, identified two immune dominant proteins from both regions, i.e.…”

“…The Treg cytokine IL-10 is associated with deactivation of macrophages and downregulates the production and nullifies the effect of the protective Th1 cytokine IFN-γ [8]. In addition, the ratio of Th1:Th2 and Th1: Treg cytokines is very important in the protection against TB because the protection is better if the ratio is biased towards Th1 cytokines [9,10].…”
