Characterization of HCV Genotype 5a Envelope Proteins: Implications for Vaccine Development and Therapeutic Entry Target

Abstract: Background:Hepatitis C virus (HCV) is one of the major causes of cirrhosis and hepatocellular carcinoma with an estimation of 185 million people with infection. The E2 is the main target for neutralizing antibody responses and the variation of this region is related to maintenance of persistent infection by emerging escape variants and subsequent development of chronic infection. While both E1 and E2 are hypervariable in nature, it is difficult to design vaccines or therapeutic drugs against them. Objectives:Th… Show more

“…Phosphorylation information is thus, critical for understanding virus protein functionality and in drug design. Prediction of this post translational modification for viral proteins has been attempted previously for Ebola virus nucleoprotein (Yasri and Wiwanitkit, 2015), envelope 2 protein of Hepatitis C virus genotype 1a (Afzal et al, 2011), envelope proteins of Hepatitis C genotype 5a (Gededzha et al, 2014), Hepatitis G virus envelope glycoprotein E2 (Ranjbar et al, 2013), whole proteome of rotavirus (Chattopadhyay et al, 2010), using computational webservers.…”

“…The method was applied to bovine ribonuclease, the B-subunit of cholera toxin and herpes simplex virus type 1 glycoprotein D. There was a good correlation between the predicted regions and previously determined antigenic regions. 10,11 The most important point of this study is starting from scratch depending on a row data from the trusty and proved sources like NCBI. All of sequences and programs that mentioned before were examined by EMBOSS web servers then decided to choose the protein sequences that produced as antibodies by the B-cell which associated with Brucellosis resistance in cattle.…”
