Characterization of Extended-Host-Range Pseudo-T-Even Bacteriophage Kpp95 Isolated on Klebsiella pneumoniae

Abstract: Kpp95, isolated on Klebsiella pneumoniae, is a bacteriophage with the morphology of T4-type phages and is capable of rapid lysis of host cells. Its double-stranded genomic DNA (ca. 175 kb, estimated by pulsed-field gel electrophoresis) can be cut only by restriction endonucleases with a cleavage site flanked either by A and T or by T, as tested, suggesting that it contains the modified derivative(s) of G and/or C. Over 26 protein bands were visualized upon sodium dodecyl sulfate-polyacrylamide gel electrophore… Show more

“…10 8 PFU in 5 l) was spotted onto lawns comprising H. pylori clinical isolates (46 from our collection plus 2 reference strains, NCTC 11637 and 26695), 10 A. baumannii strains (30), 10 E. coli strains, 6 K. pneumoniae strains, and 3 P. aeruginosa strains (51). Clearing zones formed on all H. pylori isolates but none of the other tested bacteria, suggesting that the host range of 1961P is restricted to H. pylori strains.…”

“…Many reports have reported promiscuity in enterobacterial phages (34,35,60,67,68), and MAM1 is another noteworthy example. MAM1 was found to infect 68% of the Serratia marcescens strains tested and a rhizobacterial strain from the Kluyvera genus.…”

“…Following centrifugation at 13,000 g for 30 s, the pelleted cells were resuspended in 5 ml preheated Luria-Bertani broth, followed by incubation at 37 ° . Samples were taken at 10-min intervals (up to 2 h) and immediately titered by PFU counting assay [13] . Experiments were repeated three times with duplicate samples.…”
