Characterization of E-selectin expression in vivo with use of a radiolabeled monoclonal antibody

Abstract: We have studied endothelial luminal surface expression of E-selectin in vivo in the pig. Intravenous interleukin-1 (IL-1, 5-micrograms/kg bolus +/- 50-ng.kg-1 x min-1 infusion for 2 h) induced E-selectin expression in many organs, as shown by immunostaining and selective clearance of intravenous 111In- or 99mTc-labeled anti-E-selectin monoclonal antibody (MAb 1.2B6) compared with radiolabeled immunoglobulin G1 control. Specific clearance of MAb 1.2B6 commenced 30-45 min after intravenous IL-1. Skin sites injec… Show more

“…In vivo assessment of P-selectin expression was determined using the method described by Keelan et al [25] and modified by Panes et al [26]. The monoclonal antibody RB40.34, a rat immunoglobulin G1 directed against mouse P-selectin (PharMingen, Inc.), was used for the in vivo assessment of P-selectin.…”

“…P-selectin was previously shown to be constitutively expressed on lung endothelium 14 and E-selectin on cutaneous endothelium. 15 However, in response to inflammation, both are up-regulated on endothelial cells regardless of tissue location. 13 The carbohydrate ligands for E-and P-selectins are expressed on various leukocyte subsets, including activated T cells.…”
