Characterization of complementary DNA encoding the rat neuromedin U precursor.

Abstract: Neuromedin U (NmU), a peptide originally isolated from porcine spinal cord, is known for its ability to stimulate uterine smooth muscle contraction and to cause selective vasoconstriction. It was subsequently isolated from a number of species. Among the species studied, the five amino acids at the C-terminus of the peptide are totally conserved, suggesting that this region is of major importance. We have cloned and sequenced the cDNA encoding the rat NmU precursor protein using the anchor polymerase chain reac… Show more

“…2 and 3). This is concordant with early immunological studies that characterized NmU as a gutbrain peptide and is also in agreement with Northern blot studies showing mRNA expression throughout the rat (28) and human (25) intestine and in rat pituitary (29). NmU mRNA was also observed in several regions which had previously not been studied, such as adipose tissue, again supporting a putative role of this ligand/receptor pair in modulation of energy expenditure.…”

“…Virtually nothing is known of the physiological roles that skin peptides play in amphibian survival; however, they may be involved in antipredation, modes of reproduction, and mating behavior or involved in control in osmoregulation (42). Establishment of these functions is difficult, due of the large variety of endogenous skin compounds, which could interact together to generate complex effects not produced by a single compound alone.…”

“…The cDNAs for rat and human NmU have been cloned, and analysis of the nucleic acid sequence suggests that NmU is produced as a 174-amino acid precursor (10,11). The precursor contains a signal peptide and several dibasic cleavage sites that give rise to a number of possible secreted peptides, including NmU, which is present near the carboxyl terminus.…”
