Characterization of Cd, Zn‐Thionein (Metallothionein) Isolated from Rat and Chicken Liver

Abstract: The preparation of pure metallothionein from Cd-pretreated rats and chicken was performed using short-time treatment with chloroformlethano1 followed by ion-exchange and gel chromatography. The protein contained 7 g-atoms metal ions per 12000 g protein. The Zn to Cd ratio was 1 :2.4 f 0.1. The Cd, Zn-thionein remained homogeneous during Sephadex G-25, 6-50 and 6-75 gel filtration, during polyacrylamide disc-gel electrophoresis and in the analytical ultracentrifuge. The high content of cysteine residues (approx… Show more

“…All available data indicate that the metal ions are bound through mercaptide linkages (3,20,43,44) and that all cysteinyl residues of the protein participate in metal coordination. The ratio of cysteinyl residues to the sum of cadmium and zinc is close to S in all forms of metallothionein examined thus far (2-5, 16, 20, 45).…”

“…They are unique in their high cysteine content (30%) and their absence of aromatic amino acids [l -31. Metallothioneins are induced by heavy metals in the livers of many species, including rat [4,5], rabbit [6], chicken [7] and fish [8]. Metallothioneins are also induced by metals in crustacea [9], mollusks [9,10], yeast [I 11, and Neurospora [12].…”
