Characterization of Catalase from Psychrotolerant Psychrobacter piscatorii T-3 Exhibiting High Catalase Activity

Abstract: A psychrotolerant bacterium, strain T-3 (identified as Psychrobacter piscatorii), that exhibited an extraordinarily high catalase activity was isolated from the drain pool of a plant that uses H2O2 as a bleaching agent. Its cell extract exhibited a catalase activity (19,700 U·mg protein−1) that was higher than that of Micrococcus luteus used for industrial catalase production. Catalase was approximately 10% of the total proteins in the cell extract of the strain. The catalase (PktA) was purified homogeneously … Show more

“…Three novel species of psychrotolerant H 2 O 2 -resistant bacteria, namely, V . rumoiensis [12]–[17], Exiguobacterium oxidotolerans [18][20] and Psychrobacter piscatorii [21][23], have been isolated and their catalases have been studied. Among these isolates, E. oxidotolerans T-2-2 T is the only Gram-positive bacterium and it exhibits high catalase activity in the extracellular fraction.…”

“…The thermostability of GWC is better than most heme catalases due to its four α-helix bundle robust structure. The half-life of purified GWC is 7.5 h at 80 • C. This result is better than most heme catalases such as those from Serratia marcescens SYBC08 (90 min, 60%, 70 • C) [33], Psychrobacter piscatorii T-3 (15 min, 50%, 60 • C) [34], Vibrio salmonicida (10 min, 0%, 60 • C) [35], Vibrio rumoiensis S-1 T (15 min, 0%, 60 • C) [36], Halomonas sp. SK1 (30 min, 0%, 55 • C) [37], Psychrobacter piscatorii (15 min, 20%, 65 • C) [38], and Geobacillus sp.…”

“…Other hydrogen-peroxide-tolerant bacteria that exhibit high catalase activity could be candidates for low-cost catalase production. Indeed, we have isolated other hydrogenperoxide-tolerant bacteria, namely, Vibrio rumoiensis S-1 T (Yumoto et al 1998(Yumoto et al , 1999(Yumoto et al , 2000 and Psychrobacter piscatorii T-3 (Kimoto et al 2008(Kimoto et al , 2012. Although their intracellular-catalase-producing abilities are very high, the extracellular catalase productivity of E. oxidotolerans T-2-2 T is higher than that of intracellular catalase (Table 1).…”

“…We have isolated H 2 O 2 -tolerant microorganisms from the wastewater of a fish processing plant that uses H 2 O 2 as the bleaching agent for fish eggs (Yumoto et al 1998(Yumoto et al , 1999(Yumoto et al , 2004(Yumoto et al , 2010Kimoto et al 2012) and have characterized the catalases from these isolates (Yumoto et al 2000;Hara et al 2007;Kimoto et al 2012). The high catalase activity detected in the supernatant of the spent culture medium will facilitate purification of the catalase by eliminating the need for a the cell disruption step to obtain intracellular catalase and a large amount of intracellular background proteins.…”
