Characterization of carp seminal plasma Wap65-2 and its participation in the testicular immune response and temperature acclimation

Abstract: Two functionally distinct isoforms of warm-temperature acclimation related 65-kDa protein (Wap65-1 and Wap65-2) with a role in the immune response are present in fish. To our knowledge, contrary to Wap65-1, Wap65-2 has neither been isolated nor functionally characterized in carp especially in reproductive system. The aim of this study was to characterize Wap65-2 and ascertain its functions in immune response and temperature acclimation within reproductive system. Wap65-2 corresponded to one of the most abundan… Show more

“…The data are expressed in arbitrary units according to the manufacturer’s instructions (Bio-Rad) and Posch et al [ 69 ]. To confirm the specificity of the antibodies used, protein bands corresponding to the detected bands were manually removed from the gels and prepared for digestion and protein identification using MALDI-TOF/TOF [ 70 ].…”

“…In case of both viral infection liver tissues were used. Bacterial infection with Aeromonas salmonicida was performed by intraperitoneal injection of R3xR8 cross as described here ( 26 ), liver, spleen, kidney and testis were used. Infections were performed at specific temperatures optimal for each pathogen: 18°C for CEV, 23°C for KHV and 25°C for A. salmonicida .…”

“…Total RNA was isolated from the tissues using the TRI reagent (Sigma) and transcribed to cDNA using 100 U Maxima Reverse Transcriptase (Thermo Fisher Scientific) as described earlier ( 7 , 24 26 ). cDNA samples were diluted 1:40 with nuclease-free water (Thermo Fisher Scientific) before qPCR analysis.…”
