Characterization of bone marrow CD4 to CD8 ratios and lymphocyte composition in adults by image analysis

Abstract: Bone marrow (BM) lymphocyte subsets are evaluated by flow cytometry or immunohistochemistry for diagnostic purposes; however, CD4:CD8 T lymphocyte ratios are often erroneously interpreted using peripheral blood ranges. There are few and no recent studies describing the composition of lymphocytes within the marrow space, or normal reference ranges. Lymphocyte subsets in cytopenic patients and hospital autopsy BM specimens were evaluated to better characterize CD4:CD8 ratios. Ten patients with a history of cytop… Show more

“…On bone marrow biopsies, lymphoid cells can present as either diffusely sca`ered throughout the acOve marrow space or in lymphoid aggregates including B cell aggregates. [55][56][57] Our imaging findings suggest that the infused cells may be sub-localizing to specific areas of the marrow rich in supporOve niches. Finally, in the animal receiving a second infusion, the localizaOon of foci did not match between infusions, and the degree of marrow homing was similar, suggesOng that doses of up to ten million cells per kilogram did not "fill" marrow or spleen niches, leaving remaining "space", relevant for clinical applicaOons.…”
