Characterization of Anamnestic T-cell Responses Induced by Conventional Vaccines against Contagious Bovine Pleuropneumonia

Abstract: A better understanding of how T1 vaccination confers immunity would facilitate the rational design of improved vaccines against contagious bovine pleuropneumonia (CBPP). We show here that mycoplasmas-induced recall proliferation and IFN-γ responses are detected in cattle that received multiple shots of T1 vaccines. These anamnestic responses were under the strict control of CD4+ T lymphocytes. Moreover, CD62L expression indicated that both CD4+ effector memory (Tem) and central memory (Tcm) T lymphocytes are e… Show more

“…However, within the month following vaccination series B, WC-fed calves had a greater number of CD4 + CD62L − CD45RO + T cells as compared with CFC-fed calves. Previous studies showed an increase in CD45RO on CD4 + T cells after vaccinations in both young and adult cattle (Stabel and Robbe-Austerman, 2011;Totte et al, 2013). The increase in CD4 + CD62L − CD45RO + T cells after vaccination series B could be attributed to enhanced antigen presentation and lymphocyte activation capabilities in WC-fed calves when compared with CFC-fed calves.…”

“…Interestingly, lower Mmm -induced recall activation of CD4+ T lymphocytes was detected in draining lymph nodes of animals chronically infected (i.e., presence of Mmm -containing sequestra in the lungs) in comparison to animals showing complete recovery [ 26 ]. Also, we have recently reported the absence of measurable T-cell responses in cattle one month after immunization with the attenuated T1/44 vaccine strain of Mmm [ 45 ]. The T1/44 strain also produces the EPS form of galactan [ 46 ] which may therefore potentially contribute to its poor T-cell immunogenicity.…”

“…44 As far as CBPP is concerned, helper T cells seem to play a primary role during the early stages of Mmm infection, greatly contributing to the production of interferon-g (IFN-g), the latter being considered protective during CBPP. 23,24,46,106,107 In contrast, depletion of CD4 þ lymphocytes did not affect the clinical outcome, the pathology, the humoral immune response, or the serum level of IFN-g after Mmm experimental infection, 89 thus raising further questions about the real significance of helper T cells in the pathogenesis of CBPP. Likewise, data about the humoral immune response after Mmm infection are few and somewhat conflicting.…”
