Characterization of Allergen (Bet v 1)-Specific T Cell Lines and Clones from Non-Allergic Individuals

Abstract: The immune response towards allergens in non-allergic healthy individuals was investigated. T cell lines (TCL) with specificity for Bet v 1, the major birch pollen allergen, were established and analysed for epitope specificity. 49 T cell clones (TCC) specific for Bet v 1 were isolated from TCLs. All TCCs revealed the Th phenotype. Cytokine production in response to specific stimulation revealed a majority of Th clones producing interleukin (IL)-4 and interferon (IFN)-γ; however, most TCCs revaled a low IL-4/I… Show more

“…During the last decade, clones specific for a variety of allergens have been described, often with heterogeneous cytokine profiles (ie. Th1, Th2, or Th0) [45][46][47][48][49][50][51][52][53]. Indeed, induction of the Th1 cytokine, IFN-γ, is common for a variety of allergens or allergen-derived peptides, irrespective of allergic status [38,44,45,47,[52][53][54][55].…”

“…Nonallergic individuals, however, produce low levels of specific IgG to allergens, a response mediated by Th0/Th1 cells. [189] The modulation of regulatory T cell subsets by adjuvant effects of S layers is a promising concept for immunotherapy and perhaps also for prophylaxis of type I allergies. In the experimental S layer vaccine recombinant birch pollen allergen Bet v 1 (rBet v 1) was conjugated to cross-linked S layers.…”

“…The sensitivity of the assays was 10 pg/ml for both IL-4 and IFN-γ. Th1/2 balance was compared by IL-4/IFN-γ as reported by Ebner et al [16]. …”
