Characterization of a pollen-specific cDNA clone from Nicotiana tabacum expressed during microgametogenesis and germination

Abstract: This report describes the isolation and characterization of a cDNA clone representing a gene specifically expressed in pollen. A cDNA library was constructed against mRNA from mature pollen of Nicotiana tabacum. It was screened differentially against cDNA from mRNA of leaf and of pollen. One clone, NTPc303, was further characterized. On northern blot this clone hybridizes to a transcript 2100 nucleotides in length. NTPc303 is abundant in pollen. Expression of the corresponding gene is restricted to pollen, bec… Show more

“…Moreover, their specific character was demonstrated by in vitro translation that presented EPPs as functional units with regulated translational activity. 33 The existence of these complexes explained the apparent discrepancy between the limitation of transcription to the first 3 h of tobacco pollen germination 29 and formation of 4 cm-long pollen tubes demanding massive translation. 48 Nevertheless, exact timing of derepression of mRNAs stored in EPP complexes remained unknown.…”

“…Moreover, the element AAATGA is repeated four times in the COPT1 promoter at positions Ϫ89, Ϫ452, Ϫ810, and Ϫ886. This element was identified in the promoter of the tobacco NTP303 and Arabidopsis AVP1 genes specifically expressed in pollen (38,39).…”

“…Genes which are transiently and specifically transcribed during pollen development are assumed to play a crucial role in the developmental processes. Summarizing, genes involved in pollen development can be classified, according to expression pattern as early [dO, 28,35], and late genes [4,7,21,32,39,42]. Additionally, genes transcribed in both stages have been isolated [ 1,24,33].…”
